UPLC法测定决明子中4种蒽醌类成分的含量 被引量:7

摘要 目的:利用UPLC法同时测定决明子中4种蒽醌类成分的含量,优化决明子中蒽醌类化合物含量测定的供试液制备方法。方法:采用Waters Acquity UPLC BEHC18(150 mm×2.1 mm,1.7μm)色谱柱;以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速为0.2 mL·min^-1,检测波长为284nm。通过单因素试验优化供试品溶液的制备方法。采用本方法和《中国药典》方法对21批决明子进行了含量测定,并作了比较。结果:决明子中橙黄决明素、大黄素、大黄酚、大黄素甲醚4种成分线性关系良好,精密度、稳定性、重复性的RSD值均小于2.0%,平均加样回收率为99.1%~105.7%。结论:所建立的UPLC方法有效、快速、简便,重现性好,可作为决明子质量控制的方法。 Objective:The contents of four anthraquinones in Cassia semen were simultaneously determined by UPLC,and the preparation method of the sample solution for the determination of anthraquinones in Cassia semen was optimized.Methods:A Waters Acquity UPLC BEHC18(150 mm×2.1 mm,1.7μm)column was used,a acetonitrile-0.1%phosphoric acid solution was used as mobile phase for gradient elution,the flow rate was 0.2 mL·min^-1,the detection wavelength was 284 nm.The preparation method of the sample solution was optimized by single factor tests.Moreover,the anthraquinone contents of Cassia semen in 21 batches were determined based on this method and Chinese Pharmacopoeia,respectively.Results:The linear relationships of aurantio-obtusin,emodin,chrysophanol and physcion in Cassia semen were good,the RSD values of precision,stability and repeatability were within 2.0%,and the recovery rates were between 99.1%and 105.7%.Conclusion:The UPLC method established in this paper is effective,rapid,simple and reproducible.It can be used for quality control of Cassia semen.
