摘要
油菜含油量主要受BnDGAT1和BnPDAT1基因调控,本研究以3组不同含油梯度的9个甘蓝型油菜为材料,利用定量PCR技术研究其在全生育期表达情况,以期从中找出导致含油量差异的主要基因。结果表明:BnDGAT1表达量在营养生长期时先上升后下降,在种子中表达量最高,在叶片、角果皮和种子中随含油量变化而变化;BnDGAT1在叶片、角果皮和种子中随含油量增加而增加;除花期外,BnDGAT1基因表达量和含油量的相关性均达极显著水平。BnPDAT1表达量在全生育期表达稳定;在幼苗期、21 d和35 d种子中达显著水平,在蕾薹期、35 d的角果皮、28 d种子中相关性均达显著水平。在叶片、角果皮和种子中,BnDGAT1的相关性高于BnPDAT1。本研究结果可为高含油材料筛选提供参考。
The oil content is regulated by BnDGAT1 and BnPDAT1 in rapeseed.In order to find out the key gene which control lipid synthesis mainly in different growth stages,three groups of nine Brassica napus with different oil content were used as raw materials to study expression regular in the whole development stage by RT-PCR.The results showed that the expression of BnDGAT1 increased at first,and then decreased during development period,until reached the highest level in seeds.The expression changed with the oil content in leaves,pericarp and seeds.There has highly significant difference between BnDGAT1 gene expression and oil content except the flowering stage.The expression of BnPDAT1 was stable during the whole growth stage.In seedling stage,21 d or35 d seeds stage,the BnPDAT1 has a significant difference,while there here has highly significant difference at bud bolting stage 35 d pericarp and 28 d seed stage.The correlation between gene expression and oil content of BnDGAT1 was higher than BnPDAT1 in leaves,pericarp and seeds.The results in this study can provide reference for screening high oil content rapeseed materials.
