摘要
目的 用RP -HPLC法测定盐酸二甲双胍的血清浓度。方法 采用Waters 2 690高效液相色谱仪 ,μBondapakC1 8柱 (1 0 μm ,2 50mm× 4 6mm) ,流动相为 0 0 1mol·L- 1 磷酸盐缓冲液 (pH 7 0 ) -乙腈 (60∶40 ) ,检测波长2 33nm ,流速 1 0ml·min- 1 。结果 盐酸二甲双胍的浓度在 0 0 79～ 3 97μg·ml- 1 范围内线性关系良好 (r=0 9999)。结论 本法操作简便 ,灵敏 ,快速 。
OBJECTIVE To develop an RP-HPLC method to determination the metformin hydrochloride concentrations in serum.METHODS A Waters 2690 HPLC instrument was used with a μBondapak C 18 column(10 μm,250 mm×4.6 mm).The mobile phase consisted of 0.01 mol·L -1 phosphate buffer(pH7.0)-acetonitrile(60∶40) with a flow rate of 1.0 ml·min -1 ,and the detective wavelength was 233 nm.RESULTS The calibration curve was linear in the range of 0.079 ～3.97 μg·ml -1 ( r = 0.9995 ).CONCLUSION The analysis method was simple,sensitive and rapid.It can be used in the pharmacokinetics and bioavailability study of metformin hydrochloride.
