摘要
目的 :研究脾虚状态时下丘脑 垂体 甲状腺轴调控功能及下丘脑、胸腺细胞核T3 受体水平变化 ,为脾虚证本质的研究提供新的实验资料。方法 :成年SD大鼠 32只随机分为 4组 (每组 8只 ) ,即正常组、脾虚组、治疗组、自复组。采用免疫放射分析法和放射免疫分析法 ,测定各组大鼠下丘脑、血清TRH、垂体、血清TSH及血清T3 、T4、FT3 、FT4、γT3 及下丘脑、胸腺细胞核T3 受体的含量。结果 :①脾虚组、自复组大鼠血清T3 、T4、FT3 、FT4以及下丘脑TRH、垂体TSH含量 ,下丘脑、胸腺细胞核T3 受体的含量均较正常组、治疗组为低 ,有极显著差异 (P <0 0 1) ;血清TRH、TSH含量均较正常组、治疗组为高 ,有极显著差异 (P <0 0 1)。②治疗组大鼠的上述指标与正常组无显著性差异 (P>0 0 5 )。结论 :脾虚状态时下丘脑、垂体、甲状腺合成、分泌及调控功能低下 ,各级反馈不能使各层次激素水平恢复正常 ;血清甲状腺激素及下丘脑、胸腺细胞核T3 受体水平的降低 ,致甲状腺激素生物效应低下 ,经由神经内分泌调节环路 ,削弱免疫功能 ,这部分反映了脾虚证的本质 ;健脾益气的四君子汤能修复下丘脑 垂体 甲状腺轴功能的损伤。
Objective:To study on the changes of hormones of hypothalamic pituitary thyroid axis,and nuclear T 3 receptors in hypothalamus cells & thymus Tlymphocytes in rat model of spleen asthenia.Methods: Rhubarb decoction was used to develop adult SD rat model of spleen asthenia.32 rats were averagely divided into four groups at random:control group、spleen asthenia group、treatment group、spontaneous rehabilitation group.The immunoradiometric assay and the radioimmunoassay were applied to measure nuclear T 3 receptors in hypothalamus cells & thymus T lymphocytes,and the hormones of hypothalamic pituitary thyroid axis of every rat.Result:①In spleen asthenia group and spontaneous rehabilitation group,rat T 3,T 4,FT 3,FT 4 serum levels,the contents of TRH hypothalamic,TSH pituitary levels,and the contents of nuclear T 3 receptors in hypothalamus cells and thymus T lymphocytes were markedly lower than those in control group,treatment group( P<0.01 );And TRH,TSH serum levels,were markedly higher than those in control group,treatment group ( P<0.01 );②The parameters between control group and treatment group had no significant difference ( P>0.05 ). Conclusion:Impaired hypothalamic pituitary thyroid regulation function may be attributed to progression of spleen asthenia.
