摘要
目的 :探讨补肾法预防呼吸道病毒感染的疗效及其作用机理。方法 :昆明种小鼠 32只 ,随机分为 4组 :空白组、模型组、补肾益肺胶囊预防组和左归丸预防组。以流感病毒鼠肺适应株(FM1)滴鼻复制小鼠病毒性肺炎模型。 2个预防组于实验第 1天至第 7天分别给予补肾益肺胶囊和左归丸 ,第 8天造模 ,第 14天处死全部小鼠。小鼠血清IL 8、TNF α采用放射免疫法测定 ,采用单因素方差检验。结果 :模型组小鼠肺指数较空白组显著升高 (P <0 0 5 ) ,预防 2组均较模型组显著下降 (P <0 0 5 )。实验各组小鼠脾指数无明显差异 (P >0 0 5 )。模型组小鼠血清IL 8、TNF α较空白组显著升高 (P <0 0 5 ) ,预防组的血清IL 8、TNF α较模型组显著下降 (P <0 0 5 )。结论 :补肾法可通过调节细胞炎症因子的表达水平 ,平衡机体免疫状态以预防和减轻病毒引起的肺部损伤。
Objective:The purpose of this study was to investigate the effect and mechanism of BUSHEN prophylactic treatment the respiratory tract infected with virus. Methods: 32 rats of the Kunming species were randomly divided into the controlled, model, BUSHENYIFEI capsule prophylaxis (BSYF) and ZUOGUI pelle prophylaxis (ZGW) groups (8/group). Rats of viral pneumonia model were replicated by dribbling nose with Influenzavirus (FM1) on day 8. Rats of the two prophylaxis groups were respectively fed with BSYF capsule (1.7g/kg.d),ZGW (2.4g/kg.d) from day 1 to day 7. All of the rats were killed at the day 14. Histopathology changes in lung were observed by HE dyeing. Interleukin 8(IL 8), tumor necrosis factor α(TNF α) in rats plasma assayed by radio immunity, lung index and spleen index, all were tested by mono agent variance analysis. Results: Lung index of the model group compared with controlled group increased significantly ( P <0 05). The two prophylaxis groups compared with model group decreased greatly ( P <0 05). There was no obvious difference of spleen index among groups ( P >0 05). By HE dyeing, there were no obvious pathological changes in the controlled group. And there were notable inflammatory changes in the model group. The degree of the changes in the prophylaxis groups was more slighter than that in the model group. And there was obvious difference between prophylaxis groups and the model group by Ridit test. Compared with control group, IL 8 and TNF α of the model group increased obviously ( P <0 05). IL 8 adn TNF α of prohylaxis groups decreased greatly contrasted with model group ( P<0 05 ). Conclusions: BUSHEN prophylaxis treatment may reduce the immunopathological injury caused by virus through regulating the expressing level of cytokine and balancing the immune state of the body.
