纯化的草鱼出血病病毒经SDS-PAGE分析,有11条多肽,即Vp1~Vp11,分子量为27×10~8~130×10~3。完整病毒颗粒与缺失核酸的病毒空壳均由相同的多肽组成。其中Vp1、Vp2、Vp7、Vp10和Vp11是主要的5个多肽,Vp1、Vp2、Vp3、Vp4、Vp5和Vp10属病毒核心成分。用90%DMSO变性的病毒基因组在兔网织红细胞裂解物翻译系统中进行体外翻译,其翻译产物经SDS-PAGE和放射自显影检查,在X感光片上有11条与病毒多肽一致的片段。此结果为呼肠孤病毒核酸具有mRNA性质增加了又一个证据。人工释放的病毒核酸,其双链RNA的一端与核心相连,构成类似反应核心的形态。核心上的12个钉状物可能是病毒基因组溢出的通道。
Purified grass carp hemorrhage virus(GCHV)was analysed by SDS-PA GE. 11 polypeptide specises with M. W. 130 ×103-27×103 were shownd clearly in the gel and five of them, Vpl,Vp2,Vp7,Vp10 and Vp11 are the major virion polypeptides. Intact and empty particles consist of the same poly peptides, of which 6,Vp1,Vp2,Vp3,Vp4,Vp5 and Vp10 belong to the virion core.The viral genome segments in 90% DMSO denatured by heating to 45℃ for 45 min were translated in the rabbit reticulocyte translation system at 37℃ for 1 hr. The translation products were analyzed in above system and 11 protein bands corresponding to the virion were displayed on the X-ray film. These results have provided another experimental evidence that the viral RNA from members of Reoviridae family serve the funtion of messenger RNA.GCHV RNA strands released and attached to cores were observed under electron microscopy. The 12 hollow spikes on the surface of the particles may be the sites from which dsRNA is extracted.
