IAA、KT、GA_3和CH对棉花胚状体发育的影响 被引量:2

摘要 棉花组织培养中所产生的胚状体在不加任何激素及CH的MS基本培养基上,部分球形胚状体可发育成具子叶的胚状体,并进而形成相当数量的正常试管苗;外加0.1mg·L(-1)的GA_3对球形胚状体及具子叶胚状体的正常发育均不利,导致大量胚状体重新形成愈伤组织;CH(500mg·L(-1))可促进球形胚状体的正常发育,大大降低异常胚状体的发生,并可抑制具子叶胚状体次生愈伤组织化,同时可使试管苗生长健壮:KT明显的促进了胚状体的正常发育,大大提高了正常具子叶胚状体及试管苗的形成率,但若同时附加一定浓度的IAA,刚抑制了KT的作用,表明IAA对胚状体的发育有害。 A number of globular shaped embryoids obtained from tissue culture of cotton(Gossypium hirsutum L)could develop into cotyledonous embroids and seedlings whenthey were cultured on MS medium without containing any phytohormone and CH. AddingGA_3(0. 1mg·L ̄(-1)had a harmful effect on the normal development process of globularshaped embryoid and cotyledonous embryoid.It could cause a lot of embrvoids to form cal -li,CH(500mg·(-1))could promote the normal development of globular shaped embry-oids and decrease the frequency of abnomal embryoid,while it could also make the regen-erated seedling strong. KT had a marked promote effect on the normal developmentp pro-cess of embryoid and greatly increased the formation percentages of normal cotyledonousembryoids and normal seedlings,however,if adding some concentration of IAA to the MS+KT medium,the normal development process was inhibited and the percentage of nor-mal cotyledonous embryoids and normal seedlings were decreased.
