摘要 建立一种较理想的体外子宫内膜立体共培养方法。方法:利用双室培养系统建立含有上皮细胞和基质细胞共培养的系统,光镜、电镜观察上皮细胞形态学变化;细胞免疫组化鉴定细胞来源。结果:扫描电镜显示,培养的上皮细胞生长汇合,可见到微绒毛细胞及局部有胞饮突的形成。透射电镜观察显示培养的上皮具有极性,表面有微绒毛,细胞间紧密连接及细胞间糖原沉积。结论:此方法可更好地模拟体内状态,是较理想的子宫内膜体外模型。 Objective: To set up an optimised three-dimensional model of endometrium in vitro. Methods: A three-dimensional model with separated endometrial epithelial and stromal cells was set up by using dual-chambered system. The cultures were observed by using light microscope, scanning electron microscope and transmission electron microscope. Immunohistochemical staining for cytokeratin and desmin were use to confirm the origin of endometrial cells. Results: Scanning electron microscope revealed a confluent epithelium with an abundance of microvilli and pinopodes at the apical surface. Transmission electron microscope indicated a polarized columnar epithelium in monolayer with basally nuclei and tight junction. Conclusion; This model represents a three-dimensional system that is alike the normal endometrium.
