内异方对子宫内膜异位症大鼠细胞因子含量、血管内皮细胞生长因子mRNA表达的影响 被引量:14

EFFECT OF NEIYI RECIPE ON CYTOKINES AND VEGF mRNA EXPRESSION LEVELS IN ENDOMETRIOTIC RATS

摘要 目的 :探讨中药内异方对子宫内膜异位症大鼠细胞因子含量及血管内皮细胞生长因子(VEGF)mRNA表达的调节作用。方法 :用CummingsAM方法复制子宫内膜异位症动物模型 ,采用ELISA法检测IL 6、IL 8、IL 10含量 ,Dotblot法检测VEGFmRNA表达水平。结果 :模型组外周血IL 6、腹腔液IL 8水平、腹腔巨噬细胞数量及巨噬细胞在脂多糖 (LPS)刺激下产生的IL 6含量显著高于正常对照组 ;内异方组异位种植内膜组织质量、腹腔巨噬细胞数量及巨噬细胞在LPS刺激下产生的IL 6水平显著低于模型组 ;模型组腹腔巨噬细胞在LPS刺激下产生的IL 10含量显著低于正常对照组和内异方组 ;模型组种植异位内膜组织VEGFmRNA表达水平明显高于内异方组。结论 :子宫内膜异位症细胞因子IL 6、IL 8、IL 10的失衡与疾病的发生发展有关 ,VEGFmRNA的过度表达与内异灶血管的形成相关 ;内异方可显著抑制异位内膜组织的生长 ,同时抑制异位灶VEGFmRNA的过度表达 ,减少异位灶血管的生成 ,并对细胞因子有明显的调节作用。 Objective:To investigate the regulating effect of Neiyi Recipe (NYF) on cytokines and vascular endothelial growth factor (VEGF) mRNA expression levels in endometriotic rats. Method: According to Cummings AM method, animal models with endometriosis was established. IL-6 ,IL-8 and IL-10 levels were determined by using double antibody sandwich enzyme-linked immunosorbent assay. The VEGF mRNA expression levels were examined by dot blot. Result: The serum IL-6 levels, peritoneal fluid IL-8 levels and peritoneal macrophage amounts were remarkably higher in model group than those in the control group, and the production of IL-6 by lipopolysaccharide(LPS)-activated peritoneal macrophages was significantly higher. The weights of endometriotic tissue and peritoneal macrophage amounts were remarkably lower in NYR treated group compared with that of untreated group, and the levels of IL-6 by LPS-activated peritoneal macrophages were significantly reduced. The LPS-stimulated IL-10 secretion in peritoneal macrophages from model group was significantly lower compared with that of control group and NYR treated group. The VEGF mRNA expression levels of endometriotic tissue were significantly higher in model group compared with those of NYR treated group. Conclusion: The abnormal of IL-6, IL-8 and IL-10 levels in endometriosis may be related to the pathogenesis of the disease. VEGF mRNA excessive expression had relation with blood vessel production of endometriotic tissue. NYR significantly restraines the growth of endometriotic tissue and VEGF mRNA excessive expression of endometriotic tissue, reduces blood vessel production of endometriotic tissue, and has remarkably regulating role in cytokines in endometriosis.
