摘要
目的 :探讨TGFβ、rhBMP - 2体外诱导外胚间充质细胞向平滑肌细胞分化的能力。 方法 :取妊娠 12 .5d胎鼠第三代下颌突外胚间充质细胞 ,分别在含 60、10 0mmol/L的TGF - β和 1.6、3 .2mmol/L的rhBMP- 2的DMEM/F12中培养。相差显微镜下观察细胞的形态变化 ,免疫组化鉴定细胞性质。结果 :培养 2d ,60pmol/L的TGF - β组细胞形态发生明显变化 ,细胞变得大而扁平 ,培养 4d ,免疫组化鉴定约 70 %的细胞分化为平滑肌细胞。rhBMP - 2组细胞形态未见明显改变 ,1.6nmol/L组免疫组化鉴定约 5 %的细胞分化为平滑肌细胞 ,未能诱导分化出神经元细胞。未加LIF的对照组细胞出现向平滑肌细胞的自然分化。结论 :TGFβ、rhBMP
AIM:To investigate the ability of TGF-β and rhBMP-2 on inducing the ectomesenchymal cells differentiating to smooth muscle cells. METHODS:60?100 mmol/L TGF-β and 1.6?3.2 mmol/L rhBMP-2 were added to the third passages of ectomesenchymal cells of mandibular process of 12.5 d fetal mice . The morphology was observed by phase-contrast microscope. Immunohistochemistry was used to identify the characteristics of cells. RESULTS:There were significant changes in the morphology of the after treated by 60 mmol/L TGFβ.The cells changed from fibroblast-like to big and flat. Immunohistochemistry showed that about 70% ectomesenchymal cells were positive to anti-α-SMA monoclonal antibody,which means that the cells had differentiated to smooth muscle cells. There were no significant changes in the morphology of the cells treated by rhBMP-2. About 5% cells in 1.6 nmol/L rhBMP-2 differentiated to smooth muscle cells. No neuron was differentiated successfully by rhBMP-2. Less than 1% control cells cultured without LIF differentiated spontaneously to smooth muscle cells. CONCLUSION:The ectomesenchymal cells could be differentiated to smooth muscle cells by TGFβ and rhBMP-2.
