毛细管区带电泳法测定地黄中梓醇的含量 被引量:17

摘要 目的:测定生地、熟地及不同炮制时间地黄中梓醇的含量,考察炮制时间对样醇含量的影响。方法:采用毛细管区带电泳内标法定量。用未徐层石英玻璃毛细管(60 cm×75μm,有效长度45 cm),检测波长210 nm,重力进样(10cm×5s),运行电压14 kV,温度20℃,背景电解质为50 mmol·L^(-1)硼砂溶液。进样前以0.5 mol·L^(-1)氢氧化钠溶液和背景电解质各冲洗3min。结果:该法平均回收率生地为98.2%,熟地为99.5%;RSD分别为2.1%和2.4%(n=5)。结论:毛细管电泳法用于测定地黄中梓醇的含量方法简便快捷,灵敏度高,重复性好。实验证明,梓醇在生地的炮制过程中随时间的延长而降低。因此,梓醇在熟地黄中含量较少,以梓醇作为熟地黄的质量指标是不妥当的。 Objective: The feasibility of catalpol quantification in Radix Rehmanniae by capillary electrophoresis was studied, and the influence of processing time on catalpol content was investigated. Method: The capillary e-lectrophoresis method with inner standard was used in quantification, the standards of the untreated fused silica capillary were as the follow: 60 cm×75μm, 45 cm effective length. The applied voltage was 14 kV and detection wavelength was 210 nm. 10 cm ×5 s gravity injection was used and the temperature was 20℃. The back ground electrolyte (BGE) was 50 mmol· L-1 borate solution. The capillary was rinsed with 0. 5 mol· L-1 sodium hydroxide and BGE for 3 minutes respectively before sample injection. Results: The average recovery of dried and prepared Rehmannia root was 98. 2% and 99. 5% , RSD was 2. 1 % and 2. 4% respectively ( n = 5). Conclusion: Capillary electrophoresis method is simple, rapid and sensitive with good reproducibility in the determination of catalpol in Radix Rehmanniae, the content of catalpol decreased while the processing time prolonged, catalpol used as the quality index of prepared Rehmannia root may be unsuitable.
