兔角膜真空冷冻干燥工艺及冻干角膜活性检测 被引量:3

Vacuum Freezedrying Process of Rabbit Corneas and the Viability Assessment of Endothelium

摘要 利用真空冷冻干燥法对家兔角膜进行冻干贮藏·给出了真空冷冻干燥角膜工艺,主要是对角膜进行四步法冷平衡,两步法速率降温,采用变幅值、变周期循环压力法实现干燥室内压力的调节·对按该工艺冻干的兔角膜进行物理法检测,均符合检测标准;进行台盼蓝、茜素红联合染色法检测,内皮细胞呈六角形镶嵌排列;进行透射电镜、扫描电镜检测,细胞微观结构完好·检测结果表明冻干角膜具有活性,按照此工艺能够冻干出适合贮藏、再植的活性角膜·对真空冷冻干燥法贮藏的兔角膜进行了移植实验,手术成功。 ?The rabbit corneas were preserved by vacuum freezedrying and the freezedrying process of corneas was presented. The freezedrying process mainly consists of the fourstep cold balance, twostep temperature drop and the pressure modulation in drying chamber by diversification cyclic pressure. The freezedried corneas were checked by physics method satisfied the examination standard. The endothelium of the freezedried corneas appear normally in morphology by the trypan blue and alizarin red; The endothelium ultrastructure of the freezedried corneas were uninjured by the transmission eletron microscope and the scanning electron microscope. The viability of the freezedrying corneas was proved by the examination methods. According to the freezedrying process, the rabbit corneas satisfies the need of cornea transplant and could be produced. The freezedrying rabbit corneas were used on animal experiment for keratoplasty in rabbit eyes, and the grafts remained clear postkeratoplasty. The vacuum freezedrying preserved corneas might be used for keratoplasty.
