湖北海棠的等位酶变异和遗传多样性研究 被引量:31

Allozymic variation and genetic diversity in Malus hupehensis (Rosaceae)

摘要 采用超薄平板微型聚丙烯酰胺等电聚焦电泳方法对湖北海棠 (Malushupehensis)的 9个野生居群和 2个人工栽培居群的等位酶变异和遗传多样性进行了初步研究。通过对 12个酶系统 2 9个酶位点的检测 ,结果表明湖北海棠有 2 5个酶位点的等位基因频率分布差异非常显著 ,有 10个居群发现稀有等位基因 ,并有 11个 (37.9% )重复位点 ;湖北海棠的遗传多样性水平很高 ,等位基因平均数A =2 .12 7、多态位点百分率P =74 .92 7、平均预期杂合度He=0 .376 ;居群间的基因分化系数GST=0 .2 2 4。与其他苹果属植物相比 ,湖北海棠具有中等丰富的遗传变异水平。居群间的基因流仅为Nm =0 .86 6 ,表明遗传漂变是影响居群遗传变异和遗传结构的一个重要因素。 Allozymic variation and genetic diversity were evaluated among nine wild and two cultivated populations of Malus hupehensis using isoelectric focusing in thin layer polyacrylamide slab gels. Twenty nine loci of 12 enzyme systems were detected and used for population genetic analysis, revealing that the distribution of allele frequencies of 25 loci were significant at P ≤0.01. Nineteen locally rare alleles were found in 10 populations sampled in the study. Of 29 loci, 11 (37.9%) were duplicate loci. A high level of genetic diversity was detected in M. hupehensis , with mean number of alleles per locus A =2.127, percentage of polymorphic loci P =74.927, and mean expected heterozygosity He =0.376. The relative magnitude of differentiation among populations is significantly higher than those of other long lived woody plants ( G ST =0.224). Compared with other Malus spp., this species has a moderate level of genetic variability. Gene flow among populations is only Nm =0.866, suggesting that gene drift plays an important role in influencing genetic variation and determining genetic structure.
