维甲酸逆转人宫颈癌耐药细胞株耐药性的实验研究 被引量:3

摘要 目的 :本研究旨在探讨全反式维甲酸 (ATRA)逆转人宫颈癌Hela/MMC耐药株对MMC的耐药作用。方法 :观察ATRA作用下Hela/MMC细胞的增殖 ,采用MTT法观察ATRA对Hela/MMC药物敏感性的影响 ,半定量RT PCR检测mdrI基因的表达情况。结果 :ATRA能提高MMC对Hela/MMC细胞的杀伤作用 ,上调细胞mdr1mRNA的表达。结论 :ATRA能逆转Hela/MMC对MMC的耐药性 。 Objective: To investigate the reversing effect of all-trans-retinoic acid(ATRA) of mitomycin C(MMC) resistance by all-trans-retinoic acid(ATRA) in drug resistant human cervical carcinoma cell line Hela/MMC.Methods:The cell proliferation was observed after treated with ATRA. The effects of ATRA on drug sensitivity in Hela/MMC cells were studied by MTT colorimetric assay.The change of c-myc mRNA levels was determined by semiquantitative RT-PCR.Results:ATRA could enhance the cytotoxicity of MMC on Hela/MMC cells.The expression levels of mdr 1 mRNA were increased in compared with that of the control after treated with ATRA.Conclusion:ATRA could reverse mitomycin C resistance of Hela/MMC cells.But this reversal mechanisms were not associated with the expression levels of mdr 1 gene.
