摘要
目的 :观察缺血预处理对缺血损伤的脊髓组织中自由基变化的影响 ,并探讨其对脊髓缺血损伤的保护作用的相关机制。方法 :将 44只健康新西兰大白兔随机分为缺血组 (2 0只 )、缺血预处理组 (2 0只 )和假手术组 (4只 )。应用腹主动脉夹闭法造成脊髓缺血模型 ,连续观察各组脊髓组织缺血后 40min ,再灌注后 2h、8h、2 4h、72h脑脊液中超氧化物歧化酶 (SOD)的活性和脊髓组织中丙二醛 (MDA)的含量 ,并记录动物后肢神经功能评分。结果 :缺血预处理组的脊髓组织中MDA含量显著低于缺血组 (P <0 .0 5) ,而脑脊液中SOD活性却明显高于缺血组 (P <0 .0 5)。缺血预处理组神经功能评分明显高于缺血组 (P <0 .0 1 )。结论 :缺血预处理具有抗氧化作用 ,能清除氧自由基 ,减少脂质过氧化的发生 ,保护细胞膜结构 ,对主动脉阻断所致的脊髓缺血损伤有良好的保护作用。
Objective: To investigate the effect of ischemic preconditioning(IPC) on the change of free radical in spinal cord injury associated with abdominal aortic cross-clamping and to explore its mechanisms. Methods: Forty-four New Zealand white rabbits were randomly divided into IPC group,ische mic group and Sham group. The concentrations of malondialdehyde(MDA) in spinal c ord and activities of superoxide dismutase(SOD) in cerebrospinal fluid were meas ured at five different time intervals,ischemic 40 min,reperfusion 2 h,reperfusio n 8 h,reperfusion 24 h,and reperfusion 72 h. Neurologic function was observed an d recorded.Results: The concentration of MDA of IPC group after reperfusion was decreased significan tly compared with that of ischemic group(P<0.05),while the activ ity of SOD was significantly increased compared with the later(P <0.05). The neurological scoring of IPC group was significantly higher than that in ischemic group(P<0.01). Conclusion: IPC has antioxidant potential,and can stabilize the cellular membrane and protec t the spinal cord from injury caused by aortic cross-clamping.
