摘要 用PBS抽提、硫酸铵分段沉淀及猪甲状腺球蛋白—Sepharose 4B亲和层析可从腰点沙克沙(phaseolus vulgaris L)中分离得沙克沙凝集素,达到电泳纯。SDS—聚丙烯酰胺凝胶电泳得出一条带,分子量为30.26×10~3u,表明该凝集素由单一亚基组成。其含糖量为5%。该凝集素能凝集兔、小鼠、鸡、人A、B、O红细胞,其凝集作用可被猪甲状腺球蛋白明显抑制。该凝集素可有效地促进淋巴细胞转化和分裂;体外实验表明,它可凝集Sp 2/0骨髓瘤细胞。 Sakesa Lectin was obtained by extraction with PBS, precipitation with ammonium sulfate, and affinity absorption on porcine thyroglobulin-Sepharose 4B from a seed of Phaseolus vulgaris L. SDS-polyacrylamide electrophoresis with or without 1% mercaptoethanol gave one band with molecular weights of 30.26×10~3U, indicating that the lectin consisted of identical subunits. Its total carbohydrate content was 5.0%. The lectin agglutinated rabbit, mouse, chicken and human ABO blood cell, and the activity of this lectin was inhibited by porcine thyroglobulin. Sakesa Lectin was a strong mitogen for human peripheral lymphocytes. The experiments in vitro showed that Sakesa Lectin could agglutinate mouse spleen lymphocytes and Sp2/0 myeloma cells.
