摘要
对侵染CMV的烟叶之绿岛进行组织培养。0.1%升汞浸泡烟草叶盘80s,能有效杀死机械伤口附近约2mm范围内叶肉细胞,造成化学伤口,叶盘接入培养基4—7天后,化学伤口褐化、干枯、愈伤组织形成能力完全被抑制。再于叶盘中央绿岛上制造微小伤口,叶盘中央出愈伤组织和绿苗的叶盘达65.8%。12%双氧水及100mg/ml卡那霉素亦有类似作用。
Mesophyll cells about 2mm around mechanical wound of the edge of leaf discs were killed and new chemical wound formed the discs were immersed in 0.1% HgCl_2 for 80 secands. The chemical wound became brown and wilt, and the forming ability of its callus was also inhibited completly after the discs were cultured for 4 to 7 days. However, callus and seedlings within leaf discs could grow wherever micromechanical wound was made. Twelve percent H_2O_2 and 100mg/ml Kanamycin had the same function.
