摘要 目的:了解三氧化二砷(As2O3)诱导的细胞凋亡是否与线粒体跨膜电位(Δψm)改变有关。方法:以Hela细胞为体外模型,应用碘化丙啶(PI)/Rhodamine123(Rh123)双重染色检测Δψm,通过测定细胞活力、流式细胞仪、HE染色及DNA电泳检测细胞凋亡。结果:As2O3处理Hela细胞后。台盼蓝拒染法测定细胞活力降低,流式细胞仪检测发现凋亡细胞明显增多,HE染色可见明显的细胞凋亡形态特征,琼脂糖凝胶电泳出现DNA梯形条带;As2O3使线粒体膜电位降低(P<0.01)。结论:As2O3在体外诱导Hela细胞凋亡的机制可能与降低线粒体膜电位有关。 Objective: To understand if apoptosis induced by arsenic trioxide(As2O3) is related to the alteration in mitochondrial tranmembrane pitentials(ΔΨm). Methods: Hela cells were used as in vitro models. theΔΨm was detected by the double staining of propidium iodide(PI) and Rhadamine 123 (Rh123), and apoptosis was confirmed by cell viability,staining of hematomylin-eosin(HE), flow cytometry as well as gel electrophoresis of genomic DNA and morphological observation. Results: After treatment of Hela cells with As2O3, cell viability observed by trypan-blue exclusion was decreased, and the number of apoptosis cells measured by flow cytometry was increased apparently, and the morphotogical characteristic of apoptosis was detected by the staining of hematomylin-eosin(HE), and DNA of treated Hela cells showed a ladder parrern and the ΔΨm was disrupted with As2O3 treatment(P<0.01). Conclusion: the mechanism of apoptosis induced by As2O3 may be related to the loss of ΔΨm.
