摘要
本文报道了中国仓鼠肺细胞(CHL)染色体畸变相应用Bt DR标记的姐妹染色单体互换(SCE),检测医用硅橡胶致突变作用。硅橡胶Eagle浸出液,剂量为400mg/ml、200mg/ml和100mg/ml;硅橡胶生理盐水浸出液,剂量为60mg/ml、40mg/ml及长×宽×厚为2.5cm×3cm×0.05cm共2块硅橡胶置于CHL细胞培养液内。结果表明,细胞与检测物接触时间,不论24h或48h,除400mg/ml组的染色体畸变为可疑阳性结果,畸变类型以断裂为主外,其它剂量组均为阴性结果。SCE法检测结果表明,除400mg/ml Eagle浸出液与正常对照的26h平均值为4.36±1.92;48h平均值为7.10±2.28相比较,有比较显著性差异(P<0.05)外,其它剂量组均为无统计学意义(P<0.05)。
This paper presents ths mutagenic effects of Eagle medium extract and Physiological saline extract of silicone rubber and 2 cubes of silicone rubber 2. 5cm×3 cm × 0.05cm) on the induction of chromosomal aberration and sister chromatid exchanges (SCE) in Chinese hamster lung (CHL) cells in vitro. The results showed that when the concentration of Eagle medium extract of silicone rubber was as high as 400mg/ml medium, the chromosomal aberration frequencies of CHL cells in 24 hrs or in 48 hrs were statistically higher than those of controls. (P<0.05). While there is no signficant difference in other treated groups as compared with those of control group. (P>0.05). The frequencies of SCE of Eagle medium extract of silicone rubber at dosage 400mg/ml group were 8.16±3.40/cell in 26 hrs and 9.96@2. 63/cell in 48 hrs. The differences between this group and control group were also significant.
