摘要 目的建立高效液相色谱-二极管阵列检测器(HPLC-DAD)法同时测定蒲公英配方颗粒中绿原酸、咖啡酸、木犀草苷、阿魏酸、菊苣酸、木樨草素的含量。方法采用HPLC-DAD法,色谱柱为Agilent C18柱(4.6 mm×150 mm,5μm),流动相为乙腈-0.2%甲酸水溶液,梯度洗脱,流速为1.0mL·min^-1,,柱温为30℃,检测波长为325 nm和350 nm。结果绿原酸、咖啡酸、木犀草苷、阿魏酸、菊苣酸、木樨草素在该条件下分离良好,分别在19.60~627.3、4.900~156.9、2.590~82.85、2.340~74.95、14.55~465.6、2.510~80.20μg·m L^-1,与峰面积线性关系良好,其精密度、重复性、稳定性和加样回收率均良好。结论该HPLC-DAD法稳定性、重复性好,可用于测定蒲公英配方颗粒中绿原酸、咖啡酸、木犀草苷、阿魏酸、菊苣酸、木樨草素的含量。 Objective To develop an high performance liquid chromatography-diode array detector(HPLC-DAD) method for simultaneous determination of chlorogenic acid, caffeic acid, luteolin, ferulic acid, chicory acid, and luteoloside in Dandelion formula granules. Methods An HPLC-DAD method was established. Separation was performed on an Agilent C18(4.6 mm×150 mm, 5 μm) with acetonitrile-0.2% formic acid water solution as the mobile phase with gradient elution. The flow rate was 1.0 mL·min^-1, the temperature of the column was 30 ℃, and the wavelenghs were 335 nm and 350 nm. Results Chlorogenic acid, caffeic acid, luteolin, ferulic acid, chicory acid, and luteolin were successfully separated with this method, with good linearity at 19.60-627.3, 4.900^-156.9, 2.590-82.85, 2.340-74.95, 14.55-465.6, and 2.510-80.20 μg·mL^-1, respectively. The precision, repeatability, stability and recovery were all good. Conclusion This HPLC-DAD method is stable and repeatable, and can be used to determine the content of chlorogenic acid, caffeic acid, luteolin, ferulic acid, chicory acid, and luteolin in Dandelion formula granules.
