摘要
目的建立高效液相色谱法-光电二极管阵列检测器(HPLC-PDA)同时测定附炎片中红景天苷、绿原酸、香蒲新苷、连翘酯苷A、异阿魏酸、菊苣酸的含量。方法采用Waters Symmetry C18色谱柱(250 mm×4.6 mm,5μm),对附炎片的甲醇提取液进行分析。流动相为乙腈-0.2%磷酸水,采用梯度洗脱法,流速为0.6~1.0 mL·min^(-1),柱温为30℃,进样量为10μL,检测波长为220 nm。结果所测6种成分在各自线性范围内与峰面积呈良好的线性关系(r为0.9997~0.9999)。平均加样回收率在98.79%~100.92%,RSD均<2.0%。红景天苷、绿原酸、香蒲新苷、连翘酯苷A、异阿魏酸、菊苣酸6个成分的含量范围分别为0.5000~0.5090、0.8627~0.8862、0.1533~0.1610、1.8668~1.9821、0.6952~0.6994、0.2653~0.2795 mg·g^(-1),其RSD值分别为1.9%、1.1%、1.8%、2.1%、0.84%、2.5%。结论所建立的方法操作简便、结果可靠,适用于附炎片的质量控制。
Objective To determine salidroside,chlorogenic acid,typhaneoside,forsythoside A,3-hydroxy-4-methoxycinnamic acid and cichoric acid in Fuyan tablets by HPLC-PDA.Methods A Waters Symmetry C18 column(250 mm×4.6 mm,5μm)was used to analyze the methanol extract of Fuyan tablets.The mobile phase was acetonitrile-0.2%phosphoric acid water,with gradient elution.The flow rate was 0.6~1.0 mL·min^(-1).The column temperature was 30℃,the injection volume was 10μL,and the detection wavelength was 220 nm.Results The 6 components had good linear relationship in their respective ranges(r at 0.9997~0.9999).The average recovery ranged 98.79%~100.92%,with the relative standard deviation(RSD)less than 2.0%.The content of salidroside,chlorogenic acid,typhaneoside,forsythoside A,3-hydroxy-4-methoxycinnamic acid and cichoric acid ranged 0.5000~0.5090,0.8627~0.8862,0.1533~0.1610,1.8668~1.9821,0.6952~0.6994,and 0.2653~0.2795 mg·g^(-1).The RSD was 1.9%,1.1%,1.8%,2.1%,0.84%,and 2.5%,respectively.Conclusion The method is easy to operate,reproducible and stable,which is suitable for the quality control of Fuyan tablets.
