土壤中一株溶磷菌的筛选和溶磷能力初探 被引量:8

Isolation and solubilization capacity characterization of a phosphate-solubilizing strain derived from soil

摘要 笔者以Ca3(PO4)2为难溶态磷酸盐,从红壤中筛选出一株具有较强溶磷能力的溶磷真菌,简称M1,遗传学分析表明该菌为黑曲霉(Aspergill usniger).M1在含Ca3(PO4)2、磷矿粉以及AlPO4的液体培养基中溶磷峰值分别为75.4,82.4,138.3mg·L^-1,存在一定的偏好性;M1液体培养基的pH和溶磷能力之间存在显著负相关关系(p≤0.01);M1的溶磷能力受碳源种类影响较大,以蔗糖和麦芽糖为碳源时的溶磷能力是以葡萄糖和果糖为碳源时的2~3倍,以淀粉和纤维素为碳源时的溶磷能力最差;通过高效液相色谱(HPLC)发现发酵液中包含乳酸、乙酸、苹果酸和柠檬酸,其中柠檬酸的含量和溶磷能力显著相关. In this study, one phosphate-solubilization fungus named M1, was isolated from red soil as it could produce transparent halos on solid media plates containing insoluble phosphate Ca 3(PO 4) 2 . Analysis based on molecular genetics showed that M1 was Aspergillus niger . The maximum soluble phosphate concentration of the supernatant of M1 cultured media containing Ca 3 (PO 4 ) 2 , AlPO 4 or phosphate rock can reach 75.4 mg·L^-1 , 138.3 mg·L^-1 and 82.4 mg·L^-1 , respectively, showing preference for AlPO 4 . Data analysis showed a significantly negative correlation ( p ≤ 0.01 ) between the soluble phosphate concentration and the corresponding pH of M1 cultured media. The phosphate solubilization capacity of M1 was strongly influenced by carbon resource forms as it got 2 to 3 times higher in sucrose or maltose offered media than that in glucose or fructose offered media and it became the lowest in starch or cellulose offered media. High performance liquid chromatography (HPLC) analysis unveiled the existence of lactic acid, acetic acid, malic acid and citric acid in fermented media, and the correlationship between phosphate-solubilization capacity and concentration of citric acid was significant.
