摘要
目的探讨膜联蛋白A1及其模拟肽Ac2-26对高糖诱导大鼠心肌细胞炎症反应的影响。方法培养大鼠心肌细胞并随机分为正常对照组(葡萄糖5.5 mmol/L)、正常干预组[葡萄糖(5.5 mmol/L)+Ac2-26 (0.1 mg/mL)]、高糖模型组(葡萄糖30 mmol/L)、高糖治疗组[葡萄糖(30 mmol/L)+Ac2-26 (0.1 mg/mL)];采用实时PCR测定心肌细胞膜联蛋白A1、磷脂酶A2 (PLA2) mRNA表达,采用酶联免疫分析法测定心肌细胞肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)水平。结果与正常对照组比较,正常干预组膜联蛋白A1、PLA2 mRNA,TNF-α、IL-1β水平下降,但差异无统计学意义(P> 0.05)。与正常对照组比较,高糖模型组膜联蛋白A1、PLA2 mRNA表达,TNF-α、IL-1β水平显著升高(P <0.01)。与高糖模型组比较,高糖治疗组PLA2 mRNA表达,TNF-α、IL-1β水平明显下降(P <0.01);但膜联蛋白A1表达下降不显著(P> 0.05)。结论膜联蛋白A1与高糖诱导大鼠心肌细胞炎症反应明显相关,Ac2-26能够明显改善高糖诱导大鼠心肌细胞的炎症反应。
Objective To study the effect of Annexin A1 and mimetic peptide Ac2-26 on myocardial inflammation induced by high glucose. Methods Rat cardiac myocytes were cultured and randomly divided into the following four groups:control(glucose 5.5 mmol/L),normal intervention [glucose(5.5 mmol/L)+Ac2-26(0.1 mg/mL)],high glucose(glucose 30 mmol/L),and high glucose treatment group[glucose(30 mmol/L)+Ac2-26(0.1 mg/mL)]. Expression of Annexin A1 and PLA2 mRNA in cardiac myocytes was measured by real-time PCR,and TNF-α and IL-1β level n was measured by enzyme-linked immunosorbent assay(ELISA). Results Annexin A1,PLA2 mRNA expression,TNF-α,and IL-1β level in the normal intervention group showed a decreasing trend compared to the normal control group.However,there was no significant difference(P > 0.05). Compared with the normal control group,Annexin A1,PLA2 mRNA expression,TNF-α,and IL-1β level increased significantly in the high glucose group(P < 0.01).Further,compared with the high glucose group,PLA2 mRNA expression,TNF-α,and IL-1β decreased significantly in the high glucose treatment group(P < 0.01),while Annexin A1 mRNA expression decreased slightly(P > 0.05). Conclusion Annexin A1 was significantly associated with high glucose-induced myocardial inflammation,and Ac2-26 significantly improved cardiomyocyte inflammation induced by high glucose inrat.
