卵巢癌患者血清HK10、DJ-1的表达及意义 被引量:17

Expression and significance of serum HK-10 and DJ-1 in patients with ovarian cancer

摘要 目的探讨卵巢癌患者血清人激肽释放酶10(HK-10)、DJ-1蛋白的表达及意义。方法选取97例卵巢癌患者(卵巢癌组),并于同期收集85例良性卵巢肿瘤患者(良性卵巢肿瘤组)和50例其他良性妇科疾病患者(对照组),采用酶联免疫吸附法检测各组血清HK-10、DJ-1水平,分析其与卵巢癌患者临床病理特征的关系,采用受试者工作特征曲线(ROC)下面积(AUC)评估血清HK-10、DJ-1对卵巢癌的诊断价值。结果卵巢癌组、良性卵巢肿瘤组血清HK-10、DJ-1水平高于对照组,且卵巢癌组高于良性卵巢肿瘤组,差异均有统计学意义(P均<0.05)。国际妇产科联盟分期Ⅲ~Ⅳ期、肿瘤细胞低分化、有淋巴结转移的卵巢癌患者血清HK-10、DJ-1水平高于Ⅰ~Ⅱ期、中高分化、无淋巴结转移者(P均<0.05)。Pearson积矩相关分析结果显示,卵巢癌患者血清HK-10与DJ-1呈正相关(r=0.728,P<0.05)。血清HK-10、DJ-1联合检测诊断卵巢癌的AUC为0.944(95%CI:0.889~0.992),灵敏度、特异度分别为0.93、0.97,准确度为0.96。结论卵巢癌患者血清HK-10、DJ-1水平升高,并与癌细胞的侵袭转移密切相关;二者联合检测有助于卵巢癌的早期诊断。 Objective To explore the expression levels and clinical significance of serum human kallikrein 10 (HK-10) and DJ-1 protein in patients with ovarian cancer. Methods Ninety-seven patients with ovarian cancer (ovarian cancer group), 85 patients with benign ovarian tumor (benign ovarian tumor group), and 50 patients with other benign gynecological diseases (control group) were collected. The serum HK-10 and DJ-1 levels between groups were detected by enzyme linked immunosorbent assay, and the relationship of HK-10 and DJ-1 with clinicopathological features of ovarian cancer patients were analyzed. The diagnostic value of serum HK-10 and DJ-1 for ovarian cancer was assessed by using AUC under ROC. Results The serum HK-10 and DJ-1 levels in the ovarian cancer group and benign ovarian tumor group were higher than those in the control group, and those in the ovarian cancer group were higher than those in the benign ovarian tumor group, with statistically significant differences (all P <0.05). The serum HK-10 and DJ-1 levels in patients with FIGO stage Ⅲ-Ⅳ, poorly differentiated tumor cells, and lymph node metastasis were higher than those in patients with stage I-Ⅱ, moderately and highly differentiated tumor cells and without lymph node metastasis (all P <0.05). Pearson product moment correlation analysis showed that the serum HK-10 was positively correlated with DJ-1 in patients with ovarian cancer ( r =0.728, P < 0.05 ). The AUC of combined detection of HK-10 and DJ-1 for ovarian cancer was 0.944 (95% CI: 0.889 -0.992), the sensitivity and specificity were 0.93 and 0.97 respectively, and the accuracy was 0.96. Conclusion The serum levels of HK-10 and DJ-1 in increase patients with ovarian cancer, which are also closely related to the invasion and metastasis of cancer cells, and the early detection of HK-10 and DJ-1 can be used as an important marker for clinical diagnosis of ovarian cancer.
