鹿茸复方对去卵巢骨质疏松大鼠骨代谢和骨钙磷含量的影响 被引量:9

Effect of Deer Antler Compound on Bone Metabolism and Osteocalcin Content in Ovariectomized Osteoporosis Rats

摘要 目的:观察鹿茸复方对去卵巢大鼠骨质疏松的影响。方法:摘除雌性Wistar大鼠卵巢构建去势大鼠骨质疏松模型,将造模成功的24只大鼠随机分为3组:雌激素组(E2组),模型组(OVX组)和鹿茸复方组(F组);另取8只手术但不去势的大鼠为假手术组(Sham组);共4组(n=8)。分别给予相应的药物灌胃,连续给药12周,处死。然后称量各组大鼠的体质量、子宫质量和股骨质量;ELISA试剂盒检测各组血清中碱性磷酸酶(ALP)、骨形成蛋白(BGP)、甲状旁腺激素(PTH)和降钙素(CT)的含量;原子吸收法测定血清钙(Ca)和血清磷(P)的含量;取左侧股骨灰化,称质量,计算灰分;EDTA法检测骨钙含量,钒钼酸铵法检测骨磷的含量;对右侧股骨进行HE染色,观察骨组织形态和结构,计算切片内骨小梁面积比值。结果:与Sham组比较,OVX组大鼠的子宫质量、脏器比值、血清CT和Ca含量、骨钙和骨磷含量、骨小梁面积比均明显降低;血清ALP、BGP、PTH、P含量和大鼠体质量增值率均显著升高。与OVX组比较,E2组大鼠血清BGP、PTH、P含量均明显降低,子宫质量和脏器比值、骨钙和骨磷含量、骨小梁面积比值均显著升高;F组大鼠的血清ALP、BGP、PTH、P含量和大鼠体质量增值率均明显降低,血清CT、子宫质量和脏器比值、股骨质量均有显著升高。与E2组比较,F组大鼠的股骨质量和血清CT均明显升高;血清ALP、BGP、PTH、P含量、子宫质量、脏器比值、大鼠体质量增值率、骨小梁面积比值显著降低。结论:以鹿茸为君药的复方可以有效通过调节大鼠体内激素分泌改善去卵巢大鼠的骨代谢异常,抑制去卵巢大鼠骨钙和骨磷矿物质含量的流失,逆转骨组织病理状态,对骨质疏松症起到一定的防治作用。 Objective:To observe the effect of antler compound on osteoporosis in ovariectomized rats.Methods:The osteoporosis model of ovariectomized rats was established by removing the ovary of female Wistar rats.The 24 successfully modeled rats were randomly divided into 3 groups:estrogen group(E2 group),model group(OVX group)and antler compound.Group(F group)of 8 rats with surgery but not castrated were sham operation group(Sham group);A total of 4 groups(n=8).The corresponding drugs were administered intragastrically,and were administered continuously for 12 weeks and sacrificed.The body weight,uterine weight and femur weight of each group of rats were then weighed.An ELISA kit was used to detect alkaline phosphatase(ALP),bone morphogenetic protein(BGP),parathyroid hormone(PTH)and calcitonin in each group.(CT)content;determination of serum calcium(Ca)and serum phosphorus(P)by atomic absorption method;taking the left femur ashing,weighing,calculating ash weight;EDTA method for detecting bone calcium content,vanadium and molybdenum method was used to detect the content of bone phosphorus;HE staining was performed on the right femur,bone morphology and structure were observed,and the ratio of trabecular area in the section was calculated.Results:Compared with Sham group,the uterus weight,organ ratio,serum CT,Ca,Ca content in femurs,P content in femurs and trabecular bone area ratio of OVX group decreased significantly,serum ALP,BGP,PTH,P content and the weight gain rate of rats increased significantly.Compared with OVX group,serum BGP,PTH and P levels in E2 group decreased significantly,uterine weight and organ ratio,Ca content infemurs,P content in femurs,and trabecular bone area ratio increased significantly,serum of group F rats ALP,BGP,PTH,P content and rat weight gain rate reduced significantly,serum CT,uterine weight and organ ratio,femur weight increased significantly.Compared with the E2 group,the femur weight and serum CT of the F group increased significantly,serum ALP,BGP,PTH,P content,uterine weight,organ ratio,and rat weight gain rate trabecular bone area ratio decreased significantly.Conclusion:The compound of antler as a medicinal herb can effectively regulates the hormone secretion in rats,improves the abnormal bone metabolism in ovariectomized rats,inhibits the loss of Ca and P content in femurs in ovariectomized rats,and reverses the pathological changes of bone tissue,and plays a certain role in prevention and treatment of osteoporosis.
