摘要 目的探讨小鼠在模拟海拔8000 m急性高原缺氧环境下不同时间点对小鼠血常规参数检测结果的影响。方法将小鼠随机分为对照组、缺氧1 day组、缺氧3 d组、缺氧5 d组、缺氧7 d组,每组10只。低压低氧动物实验舱模拟海拔8000 m高原环境,用XT-2000i动物全血细胞仪进行分析,观察血常规参数检测结果变化。结果与对照组相比,缺氧1 d后,白细胞计数(WBC)、平均红细胞血红蛋白(MCH)、血小板计数(PLT)变化显著,且观察组WBC、MCH低于对照组(P<0.05),红细胞计数(RBC)、血红蛋白浓度(HGB)、红细胞压积(HCT)变化无统计学意义(P>0.05)。与对照组相比,缺氧3、5、7 d后,WBC、RBC、HGB、HCT、MCH、PLT变化显著,且观察组MCH低于对照组,RBC、HGB、HCT高于对照组(P<0.01);观察组缺氧1、3、5 d的WBC数低于对照组,缺氧7 d的WBC数、PLT数高于对照组(P<0.01)。结论模拟急性高原缺氧不同时间小鼠的血常规指标均发生明显改变,并严重影响了小鼠的正常生理代谢,进而使机体的结构与功能发生改变。 Objective To investigate effects of simulated 8000 m altitude hypoxia environment on results of blood routine parameters at different times in mice. Methods Mice were randomly divided into control group, 1 d hypoxia group, 3 d hypoxia group, 5 d hypoxia group and 7 d hypoxia group. Hypobaric and hypoxic animal experiment chamber was used to simulate at an altitude of 8000 m plateau environment, and XT-2000i Animals Whole Blood Cell Analyzer was used to observe result changes in blood routine parameters. Results Compared with those in control group, levels of white blood cell count (WBC), mean corpuscular hemoglobin (MCH) and platelets (PLT) count were significantly different after 1 d of hypoxia, and WBC and MCH levels in observation group were significantly lower than those in control group ( P <0.05), but there were no significant differences in red blood cell count (RBC), hemoglobin concentration (HGB) and hematocrit (HCT) levels ( P >0.05). Compared with those in control group, after 3 d, 5 d and 7 d of hypoxia, levels of WBC, RBC, HGB, HCT, MCH and PLT count were significantly different;in observation group, MCH was significantly lower, while RBC, HGB and HCT levels were significantly higher than those in control group ( P < 0.01). In observation group, WBC levels after 1 d, 3 d and 5 d of hypoxia were significantly lower, while WBC and PLT count levels after 7 d of hypoxia were significantly higher than those in control group ( P <0.01). Conclusion Blood routine parameters of mice at different times under simulated acute plateau hypoxia environment are significantly changed, and they severely affect normal physiological metabolism of mice, which can further cause changes in body's structure and function.
