乳腺浸润性导管癌组织98例UbcH7表达临床意义 被引量:1

Expression of UbcH7 and its clinical significance in 98 cases of breast invasive ductal carcinoma

摘要 目的泛素交联酶L3(ubiquitin conjugating enzyme L3,UbcH7)参与了p53结合蛋白1(p53binding protein,53BP1)的蛋白酶体降解途径,且在多种恶性肿瘤中存在过表达,然而其与乳腺浸润性导管癌(invasive ductal carcinoma,IDC)的研究尚不多见。本研究旨在探讨UbcH7在IDC中的表达水平及临床意义。方法收集中国医科大学附属第一医院乳腺外科2012-01-01-2013-06-01手术切除的乳腺IDC组织及癌旁组织标本,采用免疫组化、实时荧光定量聚合酶链反应法(quantitative real-time PCR,qRT-PCR)和蛋白质印迹法检测UbcH7在IDC及癌旁组织中的表达情况,统计分析UbcH7表达与IDC患者临床病理特征及预后的关系。结果 98例乳腺IDC组织中UbcH7阳性高表达率为62.2%(61/98),高于癌旁组织的7.14%(7/98),差异有统计学意义,χ~2=32.832,P=0.000 1;UbcH7高表达与淋巴结转移(χ~2=15.675,P=0.000 1)和TNM分期(χ~2=9.188,P=0.002 6)有关联,而与患者年龄(χ~2=0.296,P=0.618 2)、组织学分级(χ~2=0.382,P=0.826 0)、肿瘤最大直径(χ~2=0.549,P=0.5309)、雌激素受体(χ~2=1.812,P=0.253 8)、孕激素受体(χ~2=0.539,P=0.526 7)、人表皮生长因子受体2(χ~2=0.078,P=0.832 4)及患者月经状态(χ~2=1.843,P=0.212 9)无关联;UbcH7高表达与低表达患者5年生存率分别为73.8%和91.9%,差异有统计学意义,χ~2=5.052,P=0.024 6。结论 UbcH7在乳腺IDC组织中表达上调,且UbcH7过表达可能参与IDC的发生、发展过程,并与侵袭转移及不良预后有关联。 OBJECTIVE Ubiquitin conjugating enzyme L3(UbcH7,also known as UBE2 L3),participated in the proteasome degradation pathway of p53 binding protein 1(53 BP1)and was overexpressed in many malignant tumors.However,studies on it and invasive ductal carcinoma of breast(IDC)were rare.The purpose of this study was to investigate the expression level and clinical significance of UbcH7 in IDC.METHDS Collected IDC tissue and paracancerous tissue samples from the department of breast surgery,the First Affiliated Hospital of China Medical University from January1 st,2012 to June 1 st,2013.Immunohistochemistry,qRT-PCR and Western Blot were used to detect the expression of UbcH7 in IDC and adjacent tissues,the relationship between UbcH7 expression and the clinicopathological characteristics and prognosis of IDC patients was statistically analyzed.RESULTS The UbcH7 positive high expression rate in 98 IDC cases was 62.2%(61/98),higher than that in the adjacent cancer tissues(7.14%)and the difference was statistically significant(χ~2=32.832,P=0.000 1).The high expression of UbcH7 was correlated with lymph node metastasis(χ~2=15.675,P=0.000 1)and TNM staging(χ~2=9.188,P=0.002 6),and was independent of the patient’s age,histological grading,tumor diameter,ER,PR,HER2,and menstrual status(χ~2=0.296,P=0.618 2;χ~2=0.382,P=0.826 0;χ~2=0.549,P=0.530 9;χ~2=1.812,P=0.253 8;χ~2=0.539,P=0.526 7;χ~2=0.078,P=0.832 4;χ~2=1.843,P=0.212 9).The 5-year survival rates of UbcH7 with high and low expression were 73.8% and 91.9% respectively(χ~2=5.052,P=0.024 6),with statistically significant differences.CONCLUSIONS UbcH7 expression is up-regulated in IDC,and UbcH7 overexpression may be involved in the occurrence and development of IDC and is related to invasion,metastasis and poor prognosis.
