摘要
目的:建立高效液相色谱法测定去炎酊中游离大黄素和大黄酚含量的方法。方法:色谱柱为Hypersil ODS(C_(18))柱(4.6 mm×250 mm,5μm),柱温为30℃;流动相为0.1%磷酸溶液-甲醇(25∶75),流速为1.0 mL/min;检测波长为254 nm。结果:大黄素和大黄酚分别在8.4604~211.5100(r=1.0000,n=6)和3.5237~88.0915(r=1.0000,n=6)μg/mL范围内呈现良好的线性关系;精密度、重复性、稳定性试验的RSD均<1.0%;平均加样回收率(RSD)分别为99.72%(0.37%)、99.78%(0.39%)。结论:该方法专属性强、简单易行、节约时间和成本,可用于测定去炎酊中游离大黄素和大黄酚的含量。
Objective To establish a high performance liquid chromatography(HPLC) method for determining the content of free emodin and chrysophanol in Quyan tincture. Method The determination was performed on Hypersil ODS(C18)(4.6 mm×250 mm,5 μm) column, and the column temperature was 30℃.The mobile phase was consisited of 0.1% phosphoric acid and methanol(25∶75).The flow rate was 1.0 mL/min. The detection wavelength was 254 nm. Result Emodin have a good linear relationship range in 8.4604~211.5100 μg/mL(r=1.000 0,n=6),and chrysophanol have a good linear relationship range in 3.5237~88.0915 μg/mL(r=1.000 0,n=6),RSD of precision、repeatability and stability test were all lower than 1.0%;The average recovery(RSD) were 99.72%(0.37%) and 99.78%(0.39%). Conclusion The method is specific, simple, time-saving and cost-saving,and can be used for the determination of free emodin and Chrysophanol in Quyan tincture.
