摘要
采用HPLC法测定清蒸大黄中的5种游离蒽醌的含量。色谱柱为Thermo BDS HYPERSII C18柱(4.6 mm×250 mm,5μm),以甲醇-0.1%磷酸(76∶24)水溶液为流动相,检测波长为254 nm,流速1.0 mL/min,柱温30℃,进样量10μL。结果表明,芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚分别在0.0207~0.4140μg (R^2=0.9999)、0.1721~3.4420μg (R^2=0.9996)、0.0203~0.4060μg (R^2=0.999 9)、0.1480~2.9600μg(R^2=0.999 8)、0.1640~3.2800μg(R^2=0.999 3)的线性范围内呈良好线性关系,说明该方法可作为大黄及其不同炮制品的质量控制方法。
To determine the contents of 5 kinds of free anthraquinones in steamed rhubarb by HPLC. The column was Thermo BDS HYPERSII C18 column(4.6 mm×250 mm,5 μm), with methanol-0.1% phosphoric acid(76∶24) aqueous solution as mobile phase, detection wavelength was 254 nm, flow rate. 1.0 m L/min, column temperature 30 ℃, injection volume 10 μL.The results showed that aloe-emodin, rhein, emodin, chrysophanol and emodin methyl ether showed good linear relationship in the linear range of 0.0207~0.4140 μg(R^2=0.9999), 0.1721~3.4420 μg(R^2=0.9996), 0.0203~0.4060 μg(R^2=0.9999),0.1480~2.9600 μg(R^2=0.9998), 0.1640~3.2800 μg(R^2=0.9993). This method can be used as a quality control method for rhubarb and its different processed products.
