摘要
目的分析hsa-miR-19a-3p与急性冠状动脉综合征(ACS)的相关性,并探讨其可能机制。方法选择2014年3月至2015年9月湖北省恩施土家族苗族自治州中心医院心血管病中心收治的118例冠心病患者作为研究对象,并按照冠心病分类分为STEMI组(34例)、NSTEMI组(25例)、UAP组(22例)和SCAD组(37例),采用实时荧光定量PCR检测血浆hsa-miR-19a-3p的表达,采用生物信息学分析其可能靶基因、生物信息学功能和可能结合非编码RNA(如miRNA、lncRNA、circRNA等);采用酶联免疫吸附法(ELISA)检测低密度脂蛋白受体相关蛋白2(LRP2)的表达。结果 STEMI组和NSTEMI组血浆hsa-miR-19a-3p水平较UAP组和SCAD组明显升高,差异具有统计学意义(P <0. 05); hsa-miR-19a-3p的靶基因可能为LRP2,其生物信息功能主要与脂蛋白生物合成过程、脂肪因子信号通路、白介素信号通路、血小板源生长因子信号通路、缺氧诱导因子激活缺氧反应、血管生成、丝/苏氨酸激酶信号通路、丝裂素活化蛋白激酶信号通路、转化生长因子β信号通路和趋化因子信号通路等有关; hsa-miR-557、hsa-miR-507、RP11-70C1. 1、CTD-2147F2. 2、C9orf106、C1orf198_hsa-circRNA8985、FGFR3_hsa-circRNA5885、CAMK2N1_hsa-circRNA8340、TGFBR3_hsa-circRNA859和BCL3_hsa-circRNA4124等可能为其"海绵";血浆LRP2水平,STEMI组(1 511±212. 9) ng/m L、NSTEMI组(1 436±400. 0) ng/m L和UAP组(587. 5±176. 0) ng/m L较SCAD组(367. 4±47. 69) ng/m L明显升高,差异具有统计学意义(P <0. 05);血浆hsa-miR-19a-3p与LRP2 Pearson相关系数为0. 488 1(95%CI:0. 312 5~0. 631 4,P=0. 000 4)。结论 hsa-miR-19a-3p与ACS密切相关,通过调控hsa-miR-19a-3p的靶基因LRP2而导致ACS的发生,可能与脂质代谢密切相关。
Objective To investigate the relationship of hsa-miR-19a-3p and acute coronary syndrome(ACS),and to explore its possible mechanism. Methods A total of 118 patients who were admitted into hospital central hospital of Enshi autonomous prefecture were selected as the objects, and they were divided into the STEMI group ( n =34),the NSTEMI group ( n =25),the UAP group ( n =22) and SCAD group ( n =37) according to the classification of coronary heart disease.The expression of hsa-miR-19a-3p was detected by the real-time fluorescence quantitative PCR. The target genes,bioinformatics function and the non-coding RNAs (miRNA,lncRNA,circRNA,etc) of hsa-miR-19a-3p′s were analyzed by the bioinformatics techniques.The expression of low density lipoprotein receptor-associated protein 2 (LRP2) was detected by enzyme-linked immunosorbent assay (ELISA). Results Plasma hsa-miR-19a-3p in STEMI group and NSTEMI group were significantly higher than the UAP group and SCAD group ( P <0.05). LRP2 may be a target gene of hsa-miR-19a-3p.And its main function of biological information may be connected with the biosynthesis of lipoprotein, fat factor signaling pathways, interleukin signaling pathways, platelet-derived growth factor signaling pathway, oxygen reaction activated by hypoxia inducing factor, angiogenesis, serine/threonine kinase signaling pathways, mitogen-activated protein kinase signaling pathways, transforming growth factor β signaling pathways and chemokine signaling pathway and so on.Hsa-miR-557,hsa-miR-507,RP11-70C1.1,CTD-2147F2.2,C9orf106,C1orf198_hsa-circRNA8985,FGFR3_hsa-circRNA5885,CAMK2N1_hsa-circRNA8340,TGFBR3_hsa-circRNA859 and BCL3_hsa-circRNA4124 may severed as its "sponge". The plasma LRP2 levels in STEMI group (1 511±212.9) ng/mL,NSTEMI group (1 436±400.0) ng/mL,UAP group (587.5±176.0) ng/mL were significantly higher than SCAD group (367.4±47.69) ng/mL,and the differences were statistically significant ( P <0.05). The Pearson correlation coefficient between plasma hsa-miR-19a-3p and LRP2 was 0.4881 (95% CI: 0.312 5~0.631 4, P =0.000 4). Conclusion Hsa-miR-19a-3p may be closely associated with ACS by regulating LRP2 through lipid metabolism process.
