摘要
目的:探讨Tg737对肝癌病灶内细胞周期进程、细胞侵袭的影响。方法:选择在本院接受手术治疗的原发性肝癌患者92例,留取术中所得肝癌组织及癌旁正常组织标本,采用荧光定量PCR检测其中Tg737基因、细胞周期相关基因、侵袭相关基因的表达量,采用Pearson检验评估肝癌组织中Tg737基因表达量与细胞周期进程、细胞侵袭的相关关系。结果:肝癌组织中Tg737mRNA的表达量显著低于癌旁组织;细胞周期相关基因cyclinA1、CDK2、PCNA mRNA的表达量均显著高于癌旁组织;侵袭相关基因CEP65、TLR2、BMPR-Ⅱ、KIF20A、Sirt1 mRNA的表达量高于癌旁组织,KAⅠ1、KLF4 mRNA的表达量低于癌旁组织。Pearson检验发现,肝癌组织中Tg737基因表达量与细胞周期相关基因、侵袭相关基因表达量直接相关。结论:肝癌病灶中Tg737基因表达量异常降低,具体降低程度与癌细胞细胞周期进程、侵袭活性直接相关。
Objective:To investigate the effects of Tg737 on cell cycle progression and cell invasion in liver cancer.Methods:92 patients with primary liver cancer who underwent surgical treatment in our hospital between January 2015 and June 2017 were selected,the liver cancer tissue and adjacent normal tissue specimens obtained during operation were kept,fluorescence quantitative PCR was adopted to detect the expression of Tg737 gene,cell cycle related genes and invasion related genes,and Pearson test was adopted to evaluate the correlation of Tg737 gene expression with cell cycle progression and cell invasion in liver cancer tissue.Results:Tg737 mRNA expression in liver cancer tissue was significantly lower than that in adjacent tissue;cell cycle related genes cyclinA1,CDK2 and PCNA mRNA expression were significantly higher than those in adjacent tissue;invasion related genes CEP65,TLR2,BMPRⅡ,KIF20A and Sirt1 mRNA expression were higher than those in adjacent tissue whereas KAⅠ1 and KLF4 mRNA expression were lower than those in adjacent tissue.Pearson test showed that the Tg737 gene expression in liver cancer tissue was directly correlated with the expression of cell cycle related genes and invasion related genes.Conclusion:Tg737 gene expression abnormally decreases in liver cancer lesion,and the specific decrease is directly correlated with cell cycle progression and invasion activity of cancer cells.
