儿童免疫性血小板减少症IL-27、CD4^+CD45RA^+T及CD4^+CD45RO^+T细胞变化 被引量:2

A study on the changes of IL-27,CD4^+CD45RA^+T and CD4^+CD45RO^+T cells in children with immune thrombocytopenia

摘要 目的检测免疫性血小板减少症患儿外周血细胞因子IL-27、CD4^+T细胞表面抗原CD45RA和CD45RO表达的变化,探讨其在ITP发病过程中的作用。方法新诊断ITP患儿35例,正常体检儿童35例作为对照,ELISA法检测血清IL-27水平、流式细胞术法检测外周血CD4^+T细胞CD45RA和CD45RO的表达率,采用t检验、直线相关分析进行比较。结果血清IL-27浓度ITP组(195.35±70.30)ng/L,对照组(301.25±125.40)ng/L,差异有统计学意义(P<0.01);CD4^+CD45RA+T细胞表达率ITP组(26.34±5.58)%,对照组(30.56±5.35)%;CD4^+CD45RO^+T细胞表达率ITP组(55.12±7.85)%,对照组(48.45±6.74)%,差异均有统计学意义(P<0.01);血清IL-27水平与外周血小板计数无相关性(r=0.202,P=0.575)。结论 IL-27、CD4^+CD45RA+T细胞和CD4^+CD45RO^+T细胞介导的细胞免疫异常可能参与了ITP发病。 Objective To explore the pathogenesis of immune thrombocytopenia(ITP)by testing the changes of expressions of IL-27,CD4c CD45RA+T and CD4+CD45RO+T in the peripheral blood in children with ITP Methods A total of35newly diagnosed ITP cases and35healthy controls were enrolled.The serum level of IL-27was detected with ELISA,the expressions of CD45RA and CD45RO in the peripheral blood CD4+T cells were detected with FCM.The results were analyzed with t test and linear correlation analysis.Results The serum expression of IL-27in the ITP group and control group was(195.35士70.30)vs(301.25士125.40)ng/L(P<0.01);the expression of CD4+CD45RA+T cells was(26.34k5.58)%vs(30.56k5.35)%(P<0.01);the expression of CD4+CD45R〇+T cells was(55.12k7.85)%vs(48.45k6.74)%(_P<0.01).There was nocorrelation between serum IL-27and platelet count(r=0.202,4=0.575).Conclusions Immune abnormalities mediated by IL-27,CD4+CD45RA+T and CD4+CD45RO+T cells may be involved in the pathogenesis of ITP.
