大白鼠肾脏发生中γ-谷氨酰转肽酶变化的免疫组织化学研究 被引量:2

IMMUNOHISTOCHEMICAL STUDY OF γ-GLUTAMYL TRANSPEPTIDASE (γ-GTP) DURING THE DEVELOPMENT OF RAT KIDNEY

摘要 应用单克隆抗体免疫组织化学方法,显示胚胎及生后大鼠肾脏的γ-谷氨酰转肽酶(γ-GTP)。从胎龄第18天起,γ-GTP在近端小管细胞刷状缘、侧面及基底面膜上反应即呈阳性,生后1~3天反应呈强阳性。随着肾单位的发育,免疫反应虽有所减弱,但生后第30天仍可看到中等强度的反应。其中近端小管S_3段的免疫反应最强,皮质深层的近端小管比皮质浅层的反应强。细段降支的立方或扁平细胞的腔面及侧面细胞膜上,免疫反应亦呈阳性。免疫电镜观察表明,生后1~7天,在近端小管上皮细胞顶部微绒毛、胞膜内陷、吞饮小泡及侧面和基底面细胞膜上,免疫反应均呈阳性,但紧密连接处反应为阴性。基底部质膜内褶的发生及γ-GTP定位,似与周围毛细血管的发生密切相关。此外,γ-GTP还存在于肾间质细胞膜上,并偶见于细胞质内。 Gamma-glutamyl transpeptidase (γ-GTP) was studied in the prenatal and neonatal rat kidneys using immunohistochemical method with monoclonal antibody to rat γ-GTP. γ-GTP was recognized on the brush border and basolateral membrane of proximal convoluted tubules from 18th embryonal day. Basolateral membrane of proximal tubules exhibited strong reaction on the 1st and 3 rd postnatal day. The reaction decreased with the development of nephrons, however, moderate reaction was still observed on 30th postnatal day in S_3 segment. Throughout the postnatal development of the kidney, S_3 segment exhibited strongest immunoreactivity and S_1 showed weakest reaction, and proximal tubules in the deep layer of the cortex always provided stronger reaction than those in the superficial layer of the cortex. Maturating cuboid and squamous epithelial cells of descending Henle's loop showed positive reaction on the luminal and lateral cell membrane. By means of immunoelectron microscopy, γ-GTP was found on the brush border, basolateral membrane, membrane of pinocytotic vesicles and apical invaginations in proximal tubular cells on the 1st-7th postnatal days, however, tight junctions were devoid of the reaction. The development of basal infolding and localization of γ-GTP seemed to be related closely to the development of surrounding capillaries. γ-GTP was localized also on the cell membrane and occasionally in the cytoplasm of interstitial cells of renal interstitium.
