摘要
目的 设计合成肿瘤抗原MAGE 12的B细胞表位抗原肽并对其进行鉴定。方法 在预测MAGE 12的B细胞表位序列为QSDEGSSNEEQE(82 -93 )的基础上 ,采用 4分支多重抗原肽结构合成抗原肽 ,免疫C 5 7BL 6小鼠 ,产生多克隆抗体 ,应用ELISA实验及免疫组化实验检测多克隆抗体是否为人的抗原肽的特异性抗体。结果 免疫C 5 7BL 6小鼠后可产生MAGE 12的抗原特异性的多克隆抗体 ,抗体滴度可达 1∶64。结论 证明所预测的表位为MAGE 12的B细胞表位。
Objective To synthesize and identify the antigenic peptide of B cell epitope on tumor antigen MAGE 12. Methods Based on predicted B cell epitope MAGE 12 82-93 , a multiple antigenic peptide (MAP) system was used to synthesize the immunogen. C 57BL/6 mice were immunized with the immunogen to produce the polyclonal anti serum. The polyclonal antibody in the serum of immunized mice was identified by using ELISA and immunohistochemical method. Results The antibody titer was reached to 1∶64. The polyclonal antibody was identified as objective specific antibody. Conclusion Those results show that the synthesized MAGE-1282-93 may be a B-cell epitope of MAGE 12.
