高晶体-高胶体渗透压混合液对脂多糖刺激的人中性粒细胞表面粘附分子CD11b表达的影响 被引量:2

The effect of hypertonic-hyperoncotic solution on CD11b expression on the lipopolysaccharide-stimulated human neutrophils in vitro

摘要 目的 了解中性粒细胞表面CD11b的表达规律,探讨高晶体-高胶体渗透压混合液(HHS)对其表达的影响。方法 分期采集10例健康人外周静脉血,Ficoll法分离中性粒细胞,LPS100ng/ml刺激,用流式细胞仪直接荧光法分析不同时段其表面CD11b表达。然后用不同浓度(0.25%,0.5%)HHS处理,分时段观察其对CD11b表达的影响。结果 脂多糖(LPS)刺激中性粒细胞15min后CD11b表达即有明显增高(P<0.01),30min达到峰值,此后再无明显变化.0.25%、0.5%高晶体-高胶体渗透压混合液均可明显抑制脂多糖诱导的CD11b表达(P<0.05),但两者之间并无明显差异(P>0.05),且其作用时间对CD11b的表达也无明显影响。结论 脂多糖刺激的人中性粒细胞表面CD11b的表达是一个快速的表达过程,半小时便可达峰值,此后维持一个恒定的表达。高晶体-高胶体渗透压混合液可明显抑制脂多糖诱导的CD11b表达,且这种作用不受HHS浓度及作用时间的影响。 Objective To investigate the effect of hypertonic-hyperoncotic solution (HHS) on CD11b expression on the surface of human neutrophils which are stimulated by lipopolysaccharide (LPS) in vitro. Methods Neutrophils were isolated from fresh peripheral venous blood of healthy volunteers aged 20-4oyr find incubated with LPS 100 ng/ml al 37℃ in 5% CO2 incubator for 15, 30, 60min, 4, 12 or 24h. HHS was prepared with 10%NaCl and 10% hydroxyethyl starch. Neutrophils were incubated with 0.25% or 0.5%HHS for 10min, 1h or 4h. Some of the HHS treated neutrophils were further subjected to LPS stimulation for 30 mm. CDllb on the surface of neutrophils were measured by using immunofluorescence and flow cytometry. Results CD11b expression on neutrophils increased significantly after being exposed to LPS for 15min (P<0.05) and reached its peak level at 30min and then remained unchanged. HHS prevented the up-regulation of CDllb expression induced by LPS. The inhibition was irrelated to the concentration of and time of exposure to HHS. Conclusion The up-regulation of CDllb expression on neutrophil induced by LPS stimulation is rapid. It can reach its peak level in a short time. HHS does not alter CD11b expression but prevents the LPS-induced up-regulation.
