摘要
目的 探讨人脑胶质瘤组织中CyclinD1和P2 7蛋白的表达及其相互关系。方法 采用SP免疫组化技术检测脑胶质瘤和非肿瘤组织中两者的表达 ,并用彩色图像分析系统作定量分析。结果 ①两者的免疫反应复合物均定位于细胞核。②两者在脑胶质瘤中的表达均显著高于非肿瘤组织 ,且CyclinD1蛋白的表达强度随肿瘤恶性程度增加而升高 ,而P2 7蛋白的表达强度则随肿瘤恶性程度增高而下降 ,两者的表达呈显著负相关。③CyclinD1蛋白的表达强度愈高则预后愈差 ,而P2 7蛋白的表达强度愈低则预后愈差。结论 两者的异常表达可能与人脑胶质瘤的发生和发展有密切关系 ,且具有协同作用 ,其表达强度可客观地反映肿瘤的恶性程度和评价预后。
Objective To study the expression of CyclinD1 and P27 protein in gliomas and their correlation. Methods Immunohistochemical technique was used to detect the expressions of CyclinD1 and P27 protein, with the result anaylzed quantitatively by using the image system. Results The immunohistochemical reaction for CyclinD1 and P27 protein was confined to the nuclei. The expressions of CyclinD1 and P27 protein in gliomas tissues of different malignant grades were remarkably different. Higher expressions of CyclinD1 or/and lower expression of P27 were associated with poor prognosis. Conclusion The abnormal expressions of both CyclinD1 and P27 might be closely related to the occurrence and development of gliomas, and they have synergetic relationship. The staining intensity of the positive nuclei of CyclinD1 and P27 protein can evaluate the prognosis and objectively reflect the malignant grade of gliomas.
