重组腺病毒介导的p27Kip1基因治疗大鼠神经胶质瘤的实验研究 被引量:2

摘要 目的研究腺病毒介导的人p27Kip1基因(Ad-p27Kip1)对大鼠神经胶质瘤生长的抑制作用。方法建立大鼠胶质瘤模型,随机分成3组:Ad-p27Kip1组、Ad-LacZ和对照组,分别给予Ad-p27Kip1、Lacz重组复制缺陷性腺病毒(Ad-LacZ)和PBS瘤内注射,观察对肿瘤生长的抑制情况,并用免疫荧光法检测p27和细胞周期素(cyclinD1)的表达;原位末端标记法检测细胞凋亡情况。结果Ad-p27Kip1组肿瘤生长受到明显抑制(P<0.05),免疫荧光染色显示移植瘤p27表达明显增强(P<0.05),cyclinD1的表达受到明显抑制(P<0.05),且细胞凋亡显著多于Ad-LacZ组和对照组(P<0.05)。结论Ad-p27Kip1对大鼠胶质瘤的生长具有明显的抑制作用,其机制可能是增强p27表达、抑制cyclinD1表达。p27Kip1有望成为胶质瘤基因治疗的新策略。 Objective To investigate the inhibitory effect of recombined adenovirus mediated human p27Kip1 gene (Ad-p27Kip1) on glioma growth in rats. Methods Twenty-four rats with gliomas caused by subcutaneous transplantation of C6 gliomas cells were divided into 3 groups, i.e. Ad-p27Kip1 treatment group, Ad-LacZ treatment group and control group. The inhibitory effect on the gliomas in all the groups was evaluated by growth curves and inhibitory rates. The expressions of p27Kip1 and cyclin D1 in the gliomas were detected by immunofluorescence technique. TdT-mediated d-UTP nick end labeling (TUNEL) was used to detect the apoptosis of glioma cells. Results The growth of tumors in Ad-p27Kip1 group was significantly suppressed (P〈0.05). The immunofluorescence assay showed that the expression of p27Kip1 was significantly increased (P〈0.05), the expression of cyclin D1 was significantly decreased (P〈0.05) in the tumors of Ad-p27Kip1 group compared to the Ad-LacZ and control groups. The apoptotic glioma cells in Adp27Kip1 group were significantly more than those in either Ad-LacZ group or control group (P〈0.05). Conclusions Ad-p27Kip1 has a significant inhibitory effect on the growth of gliomas in the rats. The inhibitory effect of Ad-p27Kip1 on the gliomas may be related to an increase in the expression of p27Kip1, and a decrease in the expression of cyclin D1. Ad-p27Kip1 gene therapy may be a promising way of treating glioma.
