摘要
本文采用硅胶柱层析法 ,从螺旋藻总脂中分离提取出硫酸喹诺糖二酰甘油酯 (SQDG) ,通过高压液相色谱 (HPL C)分离纯化得到 7个主要组分。采用气相色谱法 ,分析 SQDG中脂肪酸的含量和分子组成。结果表明 ,SQDG中主要脂肪酸为 16 :0 ,18:3n- 6 ,18:2 n- 6 ,而花生四稀酸 (2 0 :4 )及EPA(2 0 :5 )的含量要低于其他几种红藻和褐藻 ,未发现 16 :2。 SQDG中 sn- 1位 79%为不饱和脂肪酸 ,95 %以上的 sn- 2位脂肪酸为 16 :0。根据测得的 7种组分的含量和 sn- 1和 sn- 2位脂肪酸的分析计算可得 ,SQDG主要分子组成为 5 2 %~ 5 7% (18:2 ,16 :0 ) ,11% (16 :0 ,16 :0 ) ,7.1% (16 :1,16 :0 ) ,7.7% (18:1,16 :0 )
Sulphoquinovosyldiglyceride (SQDG) was separated from the total lipid of Spirulina platensis with silic gel and then was further purified into 7 main components by HPLC. Lipase XI from Rhizopus arrhizus was used to hydrolyze the fatty acids esterified in sn 1. The composition of fatty acids in total lipid and SQDG was analyzed by GC after methylation .The results show that the main fatty acids in SQDG are 16∶0,18∶3n 6,18∶2n 6. The contents of 20∶4 and 20∶5 in the SQDG of Spirulina platensis are lower than those of other red algae and brown algae while 16∶2 was undiscovered. The mole cular species analysis of SQDG shows that the unsaturated fatty acids such as C18∶2n 6 and C18∶3n 6 mainly occupy the sn 1 position, while saturated fatty acid C16:0 in the sn 2 position. The composition of SQDG is mainly 52%~57% (18∶2,16∶0), 11% ( 16∶0, 16∶0), 7.1%(16∶1,16∶0), and 7.7%(18∶1,16∶0).
