从黑曲霉发酵粉中分离纯化一种内切β-葡聚糖苷酶 被引量:4

Isolation and purification of an endoglucanase from Aspergillus niger

摘要 通过硫酸铵分级沉淀 ,CM -SephadexC - 2 5 ,DEAE -SephadexA - 5 0 ,POROS 2 0HQ离子交换色谱分离方法 ,从黑曲霉发酵粉中分离纯化了一种新的内切 β -葡聚糖苷酶 .该酶在还原条件下分子量为 39.2kD ,最适pH为 4.0 ,最适温度为 5 5℃ ,Km 为 7.41mg mL . A novel cellulase was isolated and purified to homogeneity from a commercial Aspergillus niger cellulase preparation by a combination of ammonium sulfate and three steps of ion exchange chromatograph of CM-Sephadex C-25, DEAE-Sephadex A-50 and POROS 20 HQ. The molecular weight of the purified enzyme is 39.2?kD under reduced conditions by SDS-PAGE; the optimal pH of the enzyme is 4.0, and the optimal temperature is 55?℃, its K m is 7.41?mg/mL.
