摘要
目的观察蛋氨酸对肝癌细胞和肝细胞生长的影响。方法将肝癌细胞株BEL-7402、肝细胞株L-02分别置于不含蛋氨酸而含同型半胱氨酸培养基(Met-Hcy+)和含蛋氨酸而不含同型半胱氨酸培养基(Met+Hcy-)中培养,利用细胞计数和流式细胞仪检测两组细胞增殖状态,观察蛋氨酸对各组细胞生长的影响。结果肝细胞株L-02在Met-Hcy+和Met+Hcy-中生长无差异,细胞总数及处于各细胞周期细胞数量的比例无明显差异。与Met+Hcy-组相比,在Met-Hcy+培养基中的BEL-7402细胞生长受抑,G0G1期细胞比例下降,S期细胞比例升高。结论培养基中缺少蛋氨酸,肝癌细胞株BEL-7402的生长受到抑制。
Objective To study the effect of methionine on the growth of hepatic carcinoma cell and normal hepatic cells.Methods Each of the hepatic cancer cell line BEL-7402and the hepatic cell line L-02was respectively cultured in Met - Hcy + and Met + Hcy - .The cell proliferation in different culture media was exeamined by cell counter and flow cytometry.Results There was no difference in the growth and cell cycles of the L-02cultured in Met - Hcy + and Met + Hcy - separarately.The G 0 G 1 phase percentage of BEL-7402decreased and the S phase percentage increased significantly in Met - Hcy + ,compared with that of Met + Hcy - .Conclusion The growth of hepatic carcinoma cell BEL-7402was inhabited in methionine-free medium.
