摘要
目的 :研究一氧化氮 (NO)对肺动脉平滑肌细胞 (PASMC)增殖和凋亡的作用。方法 :通过非核素标记细胞增殖检测试剂盒、3 H- Td R掺入、形态学、流式细胞术等方法研究 NO与 PASMC增殖和凋亡的关系。 结果 :2 5~ 5 0 0μm ol/L的 NO供体 S-亚硝基乙酰青霉胺 (SNAP)呈剂量依赖性地减少 PASMC的细胞数和 DNA合成 ,5 0 0μm ol/L的 SNAP显著减少 S期和G2 /M期的细胞比例 ,增加 G0 /G1 期的细胞比例。 10 0~ 5 0 0μmol/L的 SNAP呈剂量依赖性地增加 PASMC的凋亡率。结论 :NO可抑制 PASMC增殖并促进其凋亡。早期给予
Objective:To study the effect of nitric oxide(NO) on cell proliferation and apoptosis of pulmonary artery smooth muscle cells(PASMC).Methods:The relationship between NO and proliferation, apoptosis of PASMC was studied with non-isotope proliferation kit, 3H-TdR incorporation, morphology and flow cytometer. Results: NO donor SNAP(25-500 μmol/L) dose dependently decreased the cell number and the DNA synthesis of PASMC, SNAP at 500 μmol/L decreased the S and G 2/M period ratio, increased the G 0/G 1 period ratio remarkably. SNAP (100-500 μmol/L) dose dependently increased the apoptotic ratio of PASMC. Conclusion: NO decreases the proliferation and increases the apoptosis of PASMC. Giving NO at early stage may prevent vascular remodeling of pulmonary hypertension.
