摘要
目的 制备 β 萘酚辛酸酯 ,用辛酸酯酶法快速检出沙门菌。 方法 采用辛酸与氯化亚砜作用生成辛酰氯 ,再与 β 萘酚反应制得 β 萘酚辛酸酯 ;将其制成酶底物纸片 ,对底物浓度、最适 pH、反应温度及时间进行了探讨 ,确定了实验的最佳条件。 结果 本法底物最佳浓度为 2 g/L ,最适 pH7.0 ,反应温度 35℃ ,1 0 g/L固蓝B显色 ,反应时间 5min。以此条件用自制底物与进口的比较 ,效果完全一致。鉴定了标准菌株 5株 ,临床分离菌株 1 4 2株。辛酸酯酶法鉴定沙门菌的敏感性为 1 0 0 % ,特异性为94 1 %。结论 本法可代替常规检验方法提前 2~ 3d报告结果 ,为沙门菌鉴定提供了一种快速、简便。
Objective To synthesize β naphthyl caprylate as substrate for rapid detection of Salmonellaes caprylase.Methods β naphthyl caprylate was synthesized by the reaction of caprylic acid with salfoxide chloride as well as following β naphthol,and the test paper was made from it as the substrate of enzyme.The optimum concentration of substrate,pH,temperature and time of reaction,etc.were established .Results Five standard strains were correctly identified and 142 clinical isolated strains showed the sensitivity was as 100% and the specificity as 94.1%.Conclusion β naphthyl caprylate can be used for easy,rapid,accurate identification of Salmonellae .
