摘要
目的 :观察血小板源性生长因子 (PDGF)的两个亚基 (A、B)和两种受体 (PDGFRα和 PDGFRβ)在正常皮肤、溃疡边缘和溃疡组织中的分布、表达特征及其与溃疡形成的关系。方法 :2 4份被测标本来自 8例不同类型的溃疡患者 ,取其溃疡皮肤组织 8份、对应的溃疡边缘组织 8份及正常皮肤组织 8例。用常规病理技术和免疫组织化学方法确定这 4种蛋白在不同组织中的定位和表达量的变化规律。结果 :在正常皮肤组织中 ,PDGF A和 PDGF B的阳性颗粒主要见于表皮基底层细胞和内皮细胞中 ,两个亚基的阳性细胞率分别为(15 .2± 5 .6 ) %和 (2 2 .4± 7.4 ) % ;PDGFRα存在于表皮细胞和内皮细胞的细胞膜上 ,该受体表达较多 ,阳性表达率为 (36 .7± 4 .3) % ;PDGFRβ在正常皮肤中表达较弱 (15 .3± 4 .8) % ,蛋白颗粒主要集中于表皮基底层细胞膜上。从正常皮肤组织经溃疡边缘到溃疡中心 ,PDGF A和 PDGF B的蛋白表达呈升高趋势。在溃疡组织中 ,PDGF A和 PDGF B蛋白主要分布于角质形成细胞、单核细胞和巨噬细胞的胞质内 ,两者的阳性细胞率分别升至为正常皮肤的 1.4 1倍和 1.13倍。 PDGFRα主要分布于表皮基底层细胞和内皮细胞的细胞膜上 ,而含有 PDGFRβ阳性颗粒的细胞主要为表皮生发层细胞。与正常皮肤组织相比 ,α和β型受体表?
Objective:To investigate the protein expression of platelet derived growth factor (PDGF) two subunits (PDGFA,PDGFB) and their receptors,PDGFRα and PDGFRβ,in dermal chronic ulcers and normal skins in order to explore their influences on ulcer formation.Methods:The expression intensity and distribution of PDGFA,PDGFB,PDGFRα and PDGFRβ were detected with pathological method and immunohistochemistry method in 8 cases of dermal ulcers,8 cases of marginal ulcers and 8 cases of normal skins.Results:In normal skins,the positive particles of PDGFA and PDGFB could be found in epidermal basal cells and endothelial cells.The positive cellular ratios of two proteins were (15 2±5 6)% and (22 4±7 4)% respectively. PDGFRα was mainly located in the plasmalemmas of epidermal cells.The positive cellular rate was (36 7±4 3)%.Less signals of PDGFRβ could be observed in the membranes of epidermal keratinocytes and endothelial cells 〔(15 3±4 8)%〕.From normal skins,marginal ulcers to ulcers,the contents of PDGFA and PDGFB were increased progressively.In ulcerative tissues,PDGFA and PDGFB were chiefly located in keratinocytes,monocytes and macrophages.The ratios of cells which contained A and B subunits were elevated to 1 41 times and 1 13 times of normal skin levels respectively.PDGFRα was chiefly sited in cellular membrances of keratinocytes and endothelial cells.PDGFRβ was principally distributed in plasmalemmas of epidermal basal cells.When compared with normal skins,the protein expression of PDGFRα and PDGFRβ was notably decreased ( P <0 01 and P <0 05).The positive cellular ratios of two receptors were reduced to (14 2±5 2)% and (9 2±3 1)% respectively.Conclusions:The formation of dermal chronic ulcers are closely associated with the reduction of PDGFRα and PDGFRβ protein contents which lead to binding obstruction between PDGF and its receptors,although the increment of PDGF can hardly accelerated the recovery of ulcers.
