摘要
目的分析先天性小耳畸形患者长链非编码核糖核酸(longnoncodingRNA,lncRNA)和信使核糖核酸(messenger RNA,mRNA)在同一位患者的正常侧耳廓软骨和先天性小耳畸形侧耳廓软骨中的共表达网络,探讨关键lncRNA的作用。方法通过芯片数据差异表达所得出的LncRNAs和mRNAs之间的表达相关性和生物信息学分析,对影响先天性小耳畸形发生的LncRNAs和mRNAs进行了分析,经Pearson相关分析计算,Pearson相关系数绝对值大于0.99的mRNA被作为构建编码非编码基因共表达网络关系图(ConstructionofthelncRNA-mRNAco-expressionnetwork)网络关系图的目标基因。我们采用了Cytoscape(version 3.0.1)软件进行了共表达网络图的构建。结果在共表达网络图中,15个LncRNAs和120个目标靶基因一共组成了145个相关链接,其中120个正相关,25个呈负相关,一个LncRNA最多有35个靶蛋白基因与其相关联,而一个目标靶蛋白基因最多和3个LncRNAs基因相关联。结论 LncRNA-NR_028308、LncRNA-uc003jsd.1等lncRNA居于lncRNA-mRNA共表达网络的核心位置,提示其和先天性小耳畸形的发生发展有一定的关系。
Objective: To analyze the profiles of long non-coding RNA (IncRNA) -messenger RNA (mRNA) co- expression network in congenital microtia and explore the potential roles of these key lncRNAs in the development of the disease. Methods- The expression correlation and bioinformatics analysis between LncRNAs and mRNAs obtained by differential expression of microarray data were used to analyze LncRNAs and mRNAs affecting the occurrence of congenital microtia. Pearson correlation analysis showed that the mRNAs, which the absolute value of Pearson correlation coefficient were greater than 0.99, were used as target genes for constructing a network map of the coding of the IncRNA-mRNA co-expression network. We used Cytoscape (version 3.0.1 ) software to build a co-expressed network graph. Results: In the co-expression network map, 15 LncRNAs and 120 target genes formed a total of 145 related links, of which 120 were positively correlated and 25 were negatively correlated. Conclusion: The 2 key lncRNAs, IncRNA-NR_028308 and lncRNA-uc003jsd.1 may be involved in the congenital microtia.
