先天性小耳畸形差异性长链非编码RNA表达谱与生物信息学分析 被引量:4

摘要 目的探讨分析先天性小耳畸形差异性长链非编码RNA(lnc RNA)的表达谱与生物信息学分析。方法采用Arraystar高通量lnc RNAs基因芯片研究3例先天性小耳畸形的患者的小耳侧残耳软骨与正常侧耳软骨,获得lnc RNAs差异性表达谱。结果本实验所用的基因芯片包含30 586个lnc RNAs和26 109个m RNAs探针;与正常侧软骨比较,残耳软骨共检测出差异性表达Lnc RNAs共有180条,其中表达上调的Lnc RNAs有74条,表达下调的Lnc RNAs有106条。信号通路分析显示涉及到甘油酯类代谢、破骨细胞分化、肿瘤生长等。结论先天性小耳畸形存在明显的lnc RNAs差异性表达,这些lnc RNAs及某些信号通路可能在小耳畸形发生和发展中发挥重要作用。 Objective:To analyze lnc RNA expression profiles in microtia and bioinformatics analysis. Methods:Three pairs of the residual ear cartilage and normal ear cartilage of 3 patients with microtia were studied by using high-throughput lnc RNAs Arraystar gene chip,obtain lnc RNAs differential expression profiles. Results:Gene chip used in this study included 30 586 lnc RNAs and 26 109 m RNAs probe;Compared with normal cartilage,180 lnc RNAs differential expression were detected in the residual ear cartilage 74 up-regulated expression lnc RNAs and 106 down-regulated expression. Signal path analysis involves glyceride metabolism,osteoclast differentiation,tumor growth,etc. Conclusion:Microtia have obvious lnc RNAs differential expression,these lnc RNAs and certain signaling pathways may play an important role in the occurrence and development of microtia.
