摘要
目的:通过评价体外抑制人肝癌细胞(BEL-7402)活性,优化黄腐酸(FA)氧解制备工艺。方法:采用BOX-BehnKen Design法,以FA提取率及其体外抑制肿瘤细胞活性的IC50为指标,选取最优工艺。结果:应用BOX-BehnKen Design法制备FA的最佳条件为氧化剂浓度15%,氧解时间90 min,液固比3∶1。结论:BOX-BehnKen Design法稳定可靠,可用于氧解制备FA。
Objective:To optimize the preparation process of fulvic acid(FA) oxidation by evaluate the inhibition of human cell line BEL-7402 activity. Methods:Box-BehnKen design was used to optimize the technology with FA extraction rate with IC50 value(inhibiting cell line activity in vitro) as the indexes. Results: The optimal conditions for Box-Behnken design were as follow:the oxidant concentration was 15%,the extraction time was 90 minutes,and the liquid-solid ratio was 3:1. Conclusion:Box-Behnken design is stable and reliable,and can be used in the preparation of FA by oxidative degradation.
