杭白菊精油对内毒素致新西兰兔发热模型解热作用及机制研究 被引量:7

Antipyretic mechanism of Chrysanthemum morifolium essential oil on fever New Zealand rabbits model induced by endotoxin

摘要 目的探讨杭白菊精油对内毒素致新西兰兔发热模型解热作用及机制。方法采用新西兰兔发热动物模型,观察抗白菊精油对其直肠温度的影响,并采用高效液相色谱法检测其下丘脑中单胺类神经递质的含量。结果给药1h后,与模型组比较,各组肛温显著降低(P<0.05);给药2h后,与模型组比较,各组肛温极显著降低(P<0.01),给药1h,2h后,抗白菊精油各剂量组和安乃近组相比,没有显著性差异(P>0.05);杭白菊精油高剂量组、安乃近组与模型组比较,甲肾上腺素(NE)、多巴胺(DA)含量下降(P<0.05)。杭白菊精油中、高剂量组、安乃近组与模型组比较,5-羟色胺(5-HT)含量上升(P<0.05),各组与模型组比较,5-羟吲哚乙酸(5-HIAA)含量上升(P<0.05),杭白菊精油各组、安乃近组与模型组比较,3-甲氧-4-羟苯乙二醇(MHPG)含量显著下降(P<0.01)。结论抗白菊精油具有明显的解热作用,且呈剂量相关性,推断其解热作用机制可能与下丘脑组织中NE、DA、5-HT含量变化有关,使5-HT、DA、NE量上的对比关系发生变化,导致体温调定点的下降而发挥解热作用。 Objective To study the antipyretic activity and related mechanisms of essential oil from Chrysanthemum morifolium on fever New Zealand rabbits induced by endotoxin.Methods Fever New Zealand rabbit animal models which was induced by endotoxin,are used to observe the influence of Chrysanthemum morifolium essential oil on rectum temperature of New Zealand rabbit,and a high performance liquid chromatography(HPLC) was used to detect the expression of monoamide neurotransmitters in the hypothalamus tissues of New Zealand rabbits.Results After 1 h dosing,compared with the model group,the anal temperature of each group was significantly reduced.(P〈0.05).After 2 h dosing,compared with the model group,the anal temperature of each group was highly significantly reduced(P〈0.01).After 1 h and 2 h dosing,there was no significant difference between the different dosage groups of Chrysanthemum morifolium essential oil and the analgin group(P〉0.05).Compared with the model group.the content of norepinephrine(NE) and dopamine(DA) in high dosage group of Chrysanthemum morifolium essential oil and the analgin group were decreased(P〈0.05),The content of 5-hydroxytryptamine(5-ht) in high and middle dose group of Chrysanthemum morifolium essential oil and the analgin group were increased(P〈0.05),the content of 5-hydroxyindoleacetic acid(5-HIAA) in each group were increased(P〈0.05),the content of 3-methoxy-4-hydroxybenzene glycol(MHPG)in the different dosage groups of Chrysanthemum morifolium essential oil and the analgin group were decreased significantly(P〈0.01).Conclusion Chrysanthemum morifolium essential oil have obvious antipyretic effect which presented a dose-response relationship,inferred that the antipyretic mechanism of Chrysanthemum morifolium essential oil may be related with the changes of NE,DA and 5-HT contents in hypothalamus organization,the changes of contrast relationship of 5-HT,DA,NE,which led to a decline in temperature set-point and exert antipyretic effect.
