摘要
目的采用改良双侧颈总动脉结扎法建立血管性痴呆大鼠模型。方法将认知能力合格的30只Wistar大鼠随机分为假手术组和模型组,每组15只。模型组先行一侧颈总动脉结扎,3 d后再行另一侧颈总动脉结扎术;假手术组仅手术分离双侧颈总动脉不结扎。术后1周采用水迷宫实验评估大鼠学习记忆能力,记录术后30 d内大鼠的死亡情况,并于术后30 d检测两组大鼠血清及海马组织的脑源性神经营养因子(BDNF)水平。结果术后30 d内,假手术组无大鼠死亡,模型组3只(30. 0%)大鼠死亡。术后1周,两组大鼠逃避潜伏期时间均有随时间缩短的趋势,且第3、4天模型组大鼠逃避潜伏期时间长于假手术组(均P <0. 05);假手术组大鼠目的象限滞留时间长于模型组,穿越平台次数多于模型组(P <0. 05)。造模30 d后,假手术组海马组织BDNF水平高于模型组(P <0. 05)。结论成功应用改良双侧颈总动脉结扎法建立大鼠血管性痴呆模型,其可以较好模拟脑部血流量不足导致的血管性痴呆;模型大鼠的认知功能障碍可能与海马组织BDNF水平降低有关。
Objective To establish the rat model of vascular dementia by the modified bilateral common carotid artery ligation. Methods Thirty Wistar rats with qualified cognitive function were randomly divided into sham group and model group,15 cases in each group.In the model group,three days after the first ligation of unilateral common carotid artery,the second ligation was applied to the remaining lateral;the sham group received bilateral common carotid artery separation alone without ligation.Water maze test was used to assess the learning and memory ability of rats after one week of operation,the mortality of rats was recorded within 30 days after operation,and the levels of hippocampal and serum brain-derived neurotrophic factor(BDNF) of rats were detected in the two groups after 30 days of operation. Results Within 30 days after operation,no rats died in the sham group,and three rats(30.0%) died in the model group.After one week of operation,the escape latency of rats decreased with time in both groups,and the escape latency of rats on the 3rd or 4th day in the model group was longer than that in the sham group(all P 〈0.05);the rats in the sham group had longer target quadrant retention time and more platforms crossed compared with the model group( P 〈0.05).After 30 days of modeling,the level of hippocampal BDNF in the sham group was higher than that in the model group( P 〈0.05). Conclusion The rat model of vascular dementia is established by the modified bilateral common carotid artery ligation successfully,which can better simulate vascular dementia caused by cerebral blood flow insufficiency ; cognitive impairment may be associated with the decreased level of hippocampal BDNF in model rats.
