蜈蚣酶解工艺优化及抗肿瘤活性研究 被引量:5

Investigation on Optimiztion of Enzymatic Hydrolysis of Centipede and Compare Its Antitumor Activity

摘要 目的:优化蜈蚣酶解工艺并探究酶解液产物体外抗肿瘤活性。方法:采用胰蛋白酶进行酶解,以水解度为指标,在单因素实验基础上,通过响应面法优化酶解工艺。结果:蜈蚣的最佳酶解条件为酶解温度49. 6℃,酶解p H为7. 62,酶解时间7小时,加酶量3000U。在最优条件下,平均水解度为17. 77%。体外抗肿瘤试验表明蜈蚣酶解液产物具有较优增殖抑制效果,对肺腺癌A549细胞株、乳腺癌MCF-7细胞株及喉癌Hep-2细胞株的IC50值分别为1. 822mg/ml、1. 142mg/ml和1. 404mg/ml。结论:此工艺稳定可行,可应用于蜈蚣酶解工艺的优化;其抗肿瘤效果优良。 Objective: Using the degree of hydrolysis( DH) as the index,the centipede enzymatic process was optimized to investigate the antitumor activity of the product. Methods: Hydrolyzed by trypsin,on the basis of one-way ANOVA studies,the response surface analysis assay was used to obtain the best enzymatic hydrolysis process. Results: The conditions contain 3000 U of enzyme amount,hydrolyzed for 7 hours,and 7. 62 of p H at 49. 6℃. Under these conditions,the average was 17. 77 %. In vitro antitumor assay showed that the hydrolysis of centipede had a better inhibitory effect on proliferation,the IC50 of A549 cell strain,breast cancer MCF-7 cell strain,and laryngeal cancer Hep-2 cell strain were 1. 822 mg/ml,1. 142 mg/ml and 1. 404 mg/ml.Conclusion: This process is stable and feasible,and can be applied to optimize the enzymatic hydrolysis process of centipede,and its anti-tumor effect is excellent.
