ANXA2 siRNA对甲状腺未分化癌细胞凋亡及放疗敏感性的影响 被引量:5

摘要 目的:探讨膜联蛋白2(ANXA2)对甲状腺未分化癌细胞凋亡及放疗敏感性的影响。方法:将甲状腺未分化癌细胞8505c分为对照组、阴性对照组和干扰组。阴性对照组和干扰组分别转染NC siRNA和ANXA2 siRNA。采用RT-PCR和Western blot检测3组细胞ANXA2 mRNA和蛋白的表达。采用MTT法检测细胞增殖,流式细胞术检测细胞凋亡,细胞克隆实验检测放疗敏感性,Western blot检测Cleaved Caspase-3和磷酸化p38(p-p38)的表达水平。结果:与对照组和阴性对照组相比,干扰组细胞ANXA2 mRNA和蛋白表达水平及存活率降低,凋亡率升高,放疗敏感性增加,细胞中Cleaved Caspase-3、p-p38蛋白增多(P<0.05)。结论:干扰ANXA2能够抑制甲状腺未分化癌细胞增殖,促进细胞凋亡,提高放疗敏感性。 Aim: To investigate the effects of Annexin A2( ANXA2) on apoptosis and radiosensitivity of anaplastic thyroid carcinoma 8505 c cells. Methods: The 8505 c cells were divided into control group,negative control group,and transfection group. The cells in the latter 2 groups were transfected with siRNA control and ANXA2 small interfering RNA.The mRNA and protein expression levels of ANXA2 in 3 groups were detected by RT-PCR and Western blot. Then,cell proliferation was detected by MTT,cell apoptosis was detected by flow cytometry,cell cloning assay was used to evaluate radiosensitivity,and the expressions of apoptotic protein Cleaved Caspase-3 and p-p38 were detected by Western blot. Results: Compared with control group and negative control group,the expression levels of ANXA2 mRNA and protein and survival rate of cells in the transfection group decreased,the apoptosis rate and radiosensitivity increased as well as the increase of Cleaved Caspase-3 and p-p38 protein( P〈0. 05). Conclusion: Interference of ANXA2 can inhibit cell proliferation,promote cell apoptosis,and enhance radiosensitivity of undifferentiated thyroid cancer cells.
